【摘要】：目的分析云南省2015年從急性弛緩性麻痹(AFP)病例中分離到的3株柯薩奇病毒B5(CVB5)VP1區(qū)核苷酸基因特征。方法采集RD細胞和L20B細胞對AFP標本進行病毒分離,應(yīng)用反轉(zhuǎn)錄-聚合酶鏈反應(yīng)(RT-PCR)擴增病毒VP1基因,目的基因送至基因公司進行序列測定;應(yīng)用MEGA 6軟件對分離到的病毒與基因庫代表株進行進化樹分析。結(jié)果 213例AFP標本中分離到23株非脊髓灰質(zhì)炎腸道病毒(NPEV),包括3株CVB5;3株CVB5與原型株VP1區(qū)之間核苷酸(nt)相似性為80.7%~81.9%,氨基酸(aa)相似性為95.1%~96.5%;系統(tǒng)進化樹結(jié)果顯示,本研究分離的CVB5與山東分離株(JN712675)的nt相似性為91.7%~92%,與安徽分離株(KF751191)的nt相似性為92%~93%。結(jié)論云南省2015年從AFP病例中分離到的3株CVB5與山東代表株(JN712675)和安徽代表株(KF751191)相似性較高。
[Abstract]:Objective to analyze the nucleotide characteristics of three coxsackie virus (Coxsackie virus) B5(CVB5)VP1 region isolated from acute flaccid paralysis (AFP) cases in Yunnan Province in 2015. Methods AFP samples were isolated from Rd cells and L20B cells. The virus VP1 gene was amplified by reverse transcription-polymerase chain reaction (RT-PCR) and the target gene was sent to the gene company for sequencing. MEGA 6 software was used to analyze the phylogenetic tree of the virus and the representative strains of gene bank. Results Twenty-three strains of non-poliomyelitis enterovirus were isolated from 213 AFP specimens, including 3 strains of CVB5 and 3 strains of CVB5. The similarity of nucleotide sequence was 80.7% and 81.9%, and the similarity of amino acid was 95.1%. The phylogenetic tree results showed that, The NT similarity between CVB5 and Shandong strain JN712675 was 91.7, and that with Anhui isolate KF751191) was 920.The similarity of NT was 92X. Conclusion there is a high similarity between the three CVB5 strains isolated from AFP cases in Yunnan Province in 2015 and the Shandong representative strain JN712675) and the Anhui representative strain KF751191).
