文章摘要:循環(huán)腫瘤細(xì)胞(circulacing tumor cells,CTCs)是指從原發(fā)腫瘤或轉(zhuǎn)移灶脫落、發(fā)生上皮-間質(zhì)轉(zhuǎn)化進(jìn)入患者外周血血液循環(huán)的惡性腫瘤細(xì)胞.CTCs在腫瘤研究和臨床診斷上的作用逐漸得到認(rèn)可,外周血中CTCs存在與否以及數(shù)量多少不但可以用于腫瘤的早期診斷,還可以用于評(píng)估腫瘤預(yù)后、監(jiān)測(cè)腫瘤的轉(zhuǎn)移和復(fù)發(fā).微流控芯片作為一個(gè)高通量、小型化的細(xì)胞實(shí)驗(yàn)平臺(tái),已被應(yīng)用于CTCs的分選當(dāng)中.本文綜述了用于CTCs捕獲的微流控芯片系統(tǒng)的最新研究進(jìn)展,著重介紹各類芯片的捕獲原理、芯片結(jié)構(gòu)和捕獲效率,最后對(duì)微流控芯片技術(shù)在CTCs分選中的應(yīng)用前景進(jìn)行了展望.
Abstr:Circulating tumor ceils (CTCs) are shed by primary or metastatic tumors, undergo the epithelial-mesenchymal transition, and enter into the peripheral blood of patients with metastatic cancers. Its critical roles in cancer research and clinical diagnostics are gradually being recognized. The presence of CTCs, especially the number in peripheral blood can not only be used for early diagnosis of cancer but also be used to assess prognosis, and monitor tumor metastasis and recurrence. Microfluidic chip as a high-throughput, miniaturized cell experimental platform has been used in CTCs sorting. In this review, we highlight recent progresses of CTCs capturing of microfluidic chip system and focus on the capture theory, chip structure and capture efficiency of various types of chips. At last, we prospect for the application foreground of microfluidic chip technology in CTCs isolation.
