反相高效液相色譜-脈沖電化學檢測法測定重組人生長激素中異丙基-β-D-硫代半乳糖苷(英文)

【摘要】:采用反相高效液相色譜(RP-HPLC)-脈沖電化學檢測(PED)法測定了重組人生長激素(rhGH)中的誘導劑異丙基-β-D-硫代半乳糖苷(IPTG)。rhGH樣品經超濾離心后,用超純水提取,經C18色譜柱分離,用脈沖電化學器檢測。結果表明,樣品中添加0.02~0.10 mg/L的IPTG,其回收率為100%~102%,相對標準偏差(n=3)小于10%。在優(yōu)化的條件下,IPTG的檢出限可達1μg/L(0.1 pmol,25μL)。該方法簡便高效,具有良好的靈敏度、回收率和重復性。
[Abstract]:The inducer isopropyl- 尾 -D- thiogalactoside (IPTGG) in recombinant human growth hormone (rhGH) was determined by reversed-phase high performance liquid chromatography (RP-HPLC-) -pulsed electrochemical detection (PED) method. After ultrafiltration centrifugation, the samples were extracted with ultrapure water and separated by C18 column. The results of pulse electrochemical detector showed that the recovery rate of the IPTGs added 0.02 ~ 0.10 mg/L was 100 ~ 102and the relative standard deviation was less than 100.The detection limit of IPTG could reach 1 渭 g / L ~ 0.1pmol ~ (-1) ~ (25 渭 L) under the optimized conditions. The method was simple and efficient, and had good sensitivity. Recovery and repeatability.
