LPS和CpG寡聚DNA對(duì)單核巨噬細(xì)胞的效應(yīng)和作用機(jī)制的研究

革蘭氏陰性細(xì)菌胞壁外膜中的脂多糖(LPS)以及細(xì)菌DNA和含非甲基化CpG基序的寡聚脫氧核糖核苷酸(CpG ODN)等都被機(jī)體當(dāng)成危險(xiǎn)信號(hào),可激活單核巨噬細(xì)胞、樹(shù)突狀細(xì)胞等抗原提呈細(xì)胞合成和分泌多種細(xì)胞因子及其它炎癥介質(zhì)。本研究主要探討了一種新型樹(shù)突狀細(xì)胞來(lái)源的G蛋白信號(hào)轉(zhuǎn)導(dǎo)調(diào)節(jié)蛋白(DC-RGS)、核受體過(guò)氧化體增殖物激活受體(PPAR)及主要組織相容性復(fù)合物Ⅱ類分子(MHC-Ⅱ)在LPS和CpG ODN活化單核巨噬細(xì)胞等抗原提呈細(xì)胞產(chǎn)生細(xì)胞因子及一氧化氮(NO)過(guò)程中的作用。我們通過(guò)基因轉(zhuǎn)染研究發(fā)現(xiàn)DC-RGS對(duì)LPS誘導(dǎo)人單核細(xì)胞THP-1產(chǎn)生細(xì)胞因子(TNF-α、IL-1β、IL-6和IL-12p70)具有微調(diào)作用,但不影響IL-8的分泌;CpG ODN也具有與LPS相似的脫敏現(xiàn)象而且它們可以互相誘導(dǎo)對(duì)方脫敏;脫敏時(shí)THP-1分泌IL-8不受抑制;用PPAR的配體和基因轉(zhuǎn)染研究發(fā)現(xiàn)PPARα和PPARγ能以配體和轉(zhuǎn)錄活性非依賴性機(jī)制抑制巨噬細(xì)胞RAW264.7產(chǎn)生IL-12p40和NO,但不影響THP-1細(xì)胞產(chǎn)生IL-8;脫敏后PPARγ表達(dá)增高提示其可能反饋性參與LPS和Cp...
