LPS和CpG寡聚DNA對單核巨噬細胞的效應(yīng)和作用機制的研究

革蘭氏陰性細菌胞壁外膜中的脂多糖(LPS)以及細菌DNA和含非甲基化CpG基序的寡聚脫氧核糖核苷酸(CpG ODN)等都被機體當(dāng)成危險信號,可激活單核巨噬細胞、樹突狀細胞等抗原提呈細胞合成和分泌多種細胞因子及其它炎癥介質(zhì)。本研究主要探討了一種新型樹突狀細胞來源的G蛋白信號轉(zhuǎn)導(dǎo)調(diào)節(jié)蛋白(DC-RGS)、核受體過氧化體增殖物激活受體(PPAR)及主要組織相容性復(fù)合物Ⅱ類分子(MHC-Ⅱ)在LPS和CpG ODN活化單核巨噬細胞等抗原提呈細胞產(chǎn)生細胞因子及一氧化氮(NO)過程中的作用。我們通過基因轉(zhuǎn)染研究發(fā)現(xiàn)DC-RGS對LPS誘導(dǎo)人單核細胞THP-1產(chǎn)生細胞因子(TNF-α、IL-1β、IL-6和IL-12p70)具有微調(diào)作用,但不影響IL-8的分泌;CpG ODN也具有與LPS相似的脫敏現(xiàn)象而且它們可以互相誘導(dǎo)對方脫敏;脫敏時THP-1分泌IL-8不受抑制;用PPAR的配體和基因轉(zhuǎn)染研究發(fā)現(xiàn)PPARα和PPARγ能以配體和轉(zhuǎn)錄活性非依賴性機制抑制巨噬細胞RAW264.7產(chǎn)生IL-12p40和NO,但不影響THP-1細胞產(chǎn)生IL-8;脫敏后PPARγ表達增高提示其可能反饋性參與LPS和Cp...
