線粒體鈣單向轉運體在氧糖剝奪-復氧復糖SH-SY5Y細胞線粒體自噬中的作用

目的 評價線粒體鈣單向轉運體(MCU)在氧糖剝奪-復氧復糖SH-SY5Y細胞線粒體自噬中的作用。方法 離體培養(yǎng)SH-SY5Y細胞,以2×105個/ml的密度接種于96孔培養(yǎng)板,采用隨機數(shù)字表法分為4組(n=6):對照組(C組)、氧糖剝奪-復氧復糖組(OGD/R組)、氧糖剝奪-復氧復糖+MCU抑制劑組(OGD/R+Ru360組)和MCU抑制劑組(Ru360組)。C組正常培養(yǎng);OGD/R組行氧糖剝奪6 h后,復氧復糖24 h;OGD/R+Ru360組氧糖剝奪前30 min加入Ru360,終濃度為10 μmol/L,其余處理同OGD/R組;Ru360組加入Ru360,終濃度為10 μmol/L,30 min后恢復正常培養(yǎng)。于復氧復糖24 h時采用CCK-8法檢測細胞存活率;JC-1法檢測細胞線粒體膜電位(MMP);透射電鏡下觀察細胞超微結構;Western blot法檢測線粒體自噬相關蛋白p62、Tom20和Beclin-1的表達。結果 與C組比較,Ru360組各項指標差異無統(tǒng)計學意義(P>0.05),OGD/R組細胞存活率和MMP下降,Tom20和p62表達下調,Beclin-1表達...
