BAMBI在小鼠不同類型脂肪組織發(fā)育及脂肪細(xì)胞分化過(guò)程中的差異表達(dá)規(guī)律

【摘要】:目的研究BAMBI基因在小鼠不同脂肪組織發(fā)育過(guò)程中的表達(dá)規(guī)律。方法利用RT-q PCR及Western blot技術(shù)檢測(cè)BAMBI基因在小鼠不同類型脂肪組織、不同發(fā)育階段(胚胎第18天、出生第0天、第21天、第8周和第6個(gè)月)及原代前體脂肪細(xì)胞分化過(guò)程中(分化第0、3、7、11天)的差異表達(dá)情況。結(jié)果隨著脂肪組織的發(fā)育,BAMBI m RNA及蛋白的表達(dá)量逐漸降低;在發(fā)育的同一時(shí)間段,BAMBI m RNA在內(nèi)臟白色脂肪組織較皮下白色脂肪組織表達(dá)量高,在棕色脂肪組織中表達(dá)量最低;在小鼠前體脂肪細(xì)胞誘導(dǎo)分化過(guò)程中,BAMBI m RNA表達(dá)量呈時(shí)間依賴性降低。結(jié)論提示BAMBI的差異表達(dá)規(guī)律與脂肪組織類型、發(fā)育階段以及脂肪細(xì)胞分化程度密切相關(guān),其在脂肪組織發(fā)育過(guò)程中起著重要的作用。
[Abstract]:Objective to study the expression of BAMBI gene in different adipose tissues of mice. Methods RT-q PCR and Western blot techniques were used to detect the BAMBI gene in different types of adipose tissues of mice at different stages of development (18th day of embryo, day 0 of birth, day 21 of birth). At the 8th week and 6th month, the differential expression of preadipocytes was observed during the differentiation of the primary adipocytes (the 0th day of differentiation). Results with the development of adipose tissue, the expression of BAMBI m RNA and protein decreased gradually, and the expression of BAMBI m RNA in visceral white adipose tissue was higher than that in subcutaneous white adipose tissue, and the lowest in brown adipose tissue. The expression of BAMBI m RNA decreased in a time dependent manner during the differentiation of mouse precursor adipocytes. Conclusion the differential expression of BAMBI is closely related to adipose tissue type, development stage and adipocyte differentiation, which plays an important role in adipose tissue development.
