【研究背景與目的】同型半胱氨酸(Homocysteine, Hcy)誘導(dǎo)神經(jīng)細(xì)胞死亡在阿爾茨海默病(Alzheimer’s disease, AD)的病理進(jìn)展中具有重要作用。硫化氫(Hydrogen sulfide,H2S)是一種內(nèi)源性神經(jīng)保護(hù)劑,具有抗氧化、抗凋亡作用。本課題組的以往研究發(fā)現(xiàn)H2S能夠拮抗Hcy的神經(jīng)細(xì)胞毒性,但其機(jī)制尚未闡明。腦源性神經(jīng)營養(yǎng)因子(Brain derived neurophic factor, BDNF)是由神經(jīng)元分泌的,在神經(jīng)細(xì)胞的分化、存活、發(fā)育中起到重要作用。作為Sirtuins家庭中的一員,沉默信息調(diào)節(jié)因子(Silent information regulator, SIRT1)在大腦、小腦及海馬中均有表達(dá),對于調(diào)節(jié)細(xì)胞的新陳代謝、衰老等起到重要作用。因而,我們將從BDNF和SIRT1的角度探討H2S拮抗Hcy神經(jīng)毒性的機(jī)制。為此,本實(shí)驗(yàn)以側(cè)腦室注射Hcy的SD大鼠為Hcy神經(jīng)毒性的動(dòng)物模型,探討H2S對Hcy損傷海馬神經(jīng)元的拮抗作用以及BDNF/TrkB通路和SIRT1對H2S抗Hcy損傷海馬神經(jīng)元的介導(dǎo)作用!痉椒ā縏unel (Termi...
