rno-miR-124-3p在大鼠急性持續(xù)性腦缺血過程中對MCT1表達的調(diào)控作用

第一部分 MCT1在急性持續(xù)性腦缺血大鼠腦組織中的表達變化目的:觀察MCT1(monocarboxylate transporter 1,MCT1)在急性持續(xù)性腦缺血大鼠腦組織中的表達變化。方法:1.健康成年雄性SD大鼠,大鼠隨機分為假手術(shù)組(n=10)和模型組(n=50),模型組又分為5個亞組,分別于造模成功后1 h,3 h,6 h,12 h,24 h取材。2.運用線栓法制作持續(xù)性大腦中動脈栓塞(permanent middle cerebral artery occlusion,p MCAO)大鼠腦缺血模型,并進行行為學(xué)觀察。3.運用TTC染色法,觀察缺血后不同時段腦組織的缺血情況變化。4.運用免疫熒光技術(shù),激光共聚焦檢測MCT1在大鼠腦組織中的表達;運用熒光雙標法檢測MCT1與星形膠質(zhì)細胞標記物GFAP(Glial fibrillary acidic protein,GFAP)的共表達情況。5.運用Western blot技術(shù),檢測腦缺血后不同時段大鼠腦組織MCT1蛋白表達變化。結(jié)果:1.構(gòu)建p MCAO模型后大鼠的行為學(xué)變化如下:1 h內(nèi)大鼠尚未蘇醒,3 h后大鼠均已蘇醒,提起...
