血腦脊液屏障上P-糖蛋白的研究進展

【摘要】:血腦脊液屏障是由腦微血管內(nèi)皮細胞、星形膠質(zhì)細胞和周皮細胞等組成的動態(tài)生理結構,對中樞神經(jīng)系統(tǒng)內(nèi)環(huán)境穩(wěn)態(tài)的維持發(fā)揮至關重要的作用。P-糖蛋白作為血腦脊液屏障上一種常見膜蛋白,它在腦微血管內(nèi)皮細胞中大量表達,限制毒性物質(zhì)和大量治療中樞神經(jīng)系統(tǒng)的藥物進入腦內(nèi)。因此對P-糖蛋白在血腦脊液屏障中的外排轉(zhuǎn)運機制的了解是神經(jīng)系統(tǒng)疾病治療的一個突破點。該文主要對P-糖蛋白的基本結構、轉(zhuǎn)運途徑、在生理與病理情況下P-糖蛋白的調(diào)節(jié)機制以及P-糖蛋白抑制劑研究進展等進行綜述,對中樞神經(jīng)系統(tǒng)藥物研發(fā)以及提高血腦脊液屏障對治療藥物通透性研究提供參考。
[Abstract]:The blood-cerebrospinal fluid barrier is a dynamic physiological structure composed of cerebral microvascular endothelial cells, astrocytes and peri-endothelial cells, which plays an important role in the maintenance of environmental homeostasis in the central nervous system. P-glycoprotein, as a common membrane protein on the blood-cerebrospinal fluid barrier, is heavily expressed in cerebral microvascular endothelial cells, restricting toxic substances and a large number of drugs for the treatment of the central nervous system into the brain. Therefore, the understanding of the efflux and transport mechanism of P-glycoprotein in the blood-cerebrospinal fluid barrier is a breakthrough point in the treatment of nervous system diseases. In this paper, the basic structure and transport pathway of P-glycoprotein, the regulation mechanism of P-glycoprotein under physiological and pathological conditions and the research progress of P-glycoprotein inhibitor were reviewed, which provided a reference for the research and development of drugs in central nervous system and the improvement of blood-cerebrospinal fluid barrier in the treatment of drug permeability.
