PAR2和TMEM16A在神經(jīng)病理性疼痛模型大鼠背根神經(jīng)節(jié)上的表達及意義

【摘要】:目的觀察坐骨神經(jīng)慢性壓迫模型(CCI)的大鼠背根神經(jīng)節(jié)(DRG)神經(jīng)元上PAR2和TMEM16A的表達,探討其在神經(jīng)病理性疼痛中發(fā)揮的作用。方法大鼠分為假手術(shù)組(Sham)和CCI組,兩組分別檢測熱縮足反射潛伏期(TWL);采用免疫熒光和Western blot檢測大鼠DRG神經(jīng)元上PAR2和TMEM16A表達。結(jié)果與Sham組比較,CCI組術(shù)前TWL差異無統(tǒng)計學意義,術(shù)后各時間點均明顯降低且差異具有統(tǒng)計學意義(P0.01)。免疫熒光結(jié)果顯示:PAR2與TMEM16A在大鼠DRG神經(jīng)元上共存。Western blot結(jié)果顯示:與Sham組相比,CCI組術(shù)后7、14 d的PAR2和TMEM16A蛋白表達均出現(xiàn)明顯增加(P0.01),且CCI組14 d高于7 d的PAR2和TMEM16A蛋白表達(P0.05)。結(jié)論神經(jīng)病理性疼痛大鼠DRG神經(jīng)元上PAR2和TMEM16A蛋白表達水平較Sham組顯著升高,這些蛋白表達水平升高可能是導致大鼠神經(jīng)病理性疼痛的原因之一。
[Abstract]:Objective to observe the expression of PAR2 and TMEM16A in (DRG) neurons of dorsal root ganglion of rats with chronic compression of (CCI), and to explore the role of PAR2 and TMEM16A in neuropathic pain. Methods the rats were divided into (Sham) group and CCI group. The latent period of thermal contractile foot reflex (TWL); was detected by immunofluorescence and Western blot. The expression of PAR2 and TMEM16A in DRG neurons was detected by immunofluorescence and Western blot respectively. Results compared with Sham group, there was no significant difference in TWL in CCI group before operation, but significantly decreased at each time point after operation (P 0.01). The results of immunofluorescence showed that PAR2 and TMEM16A coexisted on DRG neurons of rats. Western blot results showed that the expression of PAR2 and TMEM16A protein in CCI group was significantly higher than that in Sham group on the 14th day after operation (P01), and the expression of PAR2 and TMEM16A protein in CCI group was higher than that on 7th day (P 0.05). Conclusion the expression levels of PAR2 and TMEM16A proteins in DRG neurons of rats with neuropathic pain are significantly higher than those in Sham group. The increased expression of these proteins may be one of the causes of neuropathic pain in rats.
