紅曲素對大鼠腦出血后腦保護(hù)作用機(jī)制的探討

【摘要】:目的探討紅曲素(monascin)在腦出血后腦保護(hù)作用的機(jī)制及最佳劑量。方法 2016年3月—2016年10月于山西醫(yī)科大學(xué)神經(jīng)電生理實(shí)驗(yàn)室選取雄性SD大鼠120只,隨機(jī)分為假手術(shù)組、模型組、低劑量紅曲素組、中劑量紅曲素組、高劑量紅曲素組各24只。采用Ⅳ型膠原酶定位注射制備大鼠腦出血模型,于術(shù)后1、3、7、14 d行Garcia神經(jīng)功能缺損評分,行腦水含量測定、鐵染色及PPAR-γ、Nrf2、SRA測定。結(jié)果與假手術(shù)組相比,模型組、低劑量組、中劑量組及高劑量組神經(jīng)功能缺損評分在1、3、7、14 d顯著降低(F=181.705、494.229、226.576、24.726,P0.01),且在1、3 d評分最低,腦水含量在1、3 d顯著增高(F=21.781、70.036,P0.05),鐵沉積顆粒量在3、7、14 d顯著增多(F=21.426、118.122、107.773,P0.05),且于7~14 d達(dá)高峰,PPAR-γ免疫陽性細(xì)胞數(shù)于1、3、7 d顯著增多(F=3.394、28.622、14.12,P0.05),在3~7 d達(dá)高峰,Nrf2免疫陽性細(xì)胞數(shù)于1、3、7、14 d增多(F=33.076、23.17、47.779、12.182,P0.05),SRA免疫陽性細(xì)胞數(shù)于1、3、7、14 d顯著增多(F=22.283、511.092、51.811、31.060,P0.05)。與模型組相比,高劑量組Garcia評分在3~7 d時(shí)升高(P0.05),腦含水量及鐵染色顆粒量降低(P0.05),高劑量組PPAR-γ、Nff2及SPA免疫陽性細(xì)胞數(shù)于1、3及7 d明顯升高(P0.05),3~7 d達(dá)高峰,14 d降至模型組水平。結(jié)論高劑量紅曲素可增加腦出血后PPAR-γ、Nrf2及SPA陽性細(xì)胞數(shù),減輕腦出血后腦水腫及腦鐵含量,改善腦出血后神經(jīng)功能缺損。
