下調(diào)LSINCT5對人腦膠質(zhì)瘤細胞系U251增殖、遷移和放療敏感性的影響

【摘要】:目的觀察下調(diào)LSINCT5對人腦膠質(zhì)瘤細胞系U251增殖、遷移能力和放療敏感性的影響。方法采用慢病毒轉染法構建穩(wěn)定干擾LSINCT5的細胞株(U251 sh LSINCT5細胞),得到干擾效率高的sh LSINCT5 1#組、sh LSINCT5 3#組和轉染GV248的對照組細胞進行后續(xù)實驗。采用噻唑藍比色法(MTS)觀察細胞培養(yǎng)0、24、48、72h時增殖能力變化,劃痕修復實驗觀察細胞遷移能力的變化,Western blotting法檢測細胞中細胞黏附分子(E-cadherin)、緊密連接蛋白-1(Zo-1)、細胞骨架蛋白(Vimentin)表達,MTS法觀察X射線儀下分別照射0、2、4、8 Gy后細胞增殖能力。結果 sh LSINCT5 1#組、sh LSINCT5 3#組培養(yǎng)48、72 h時細胞增殖能力均較對照組低(P均0.05)。sh LSINCT5 1#組、sh LSINCT5 3#組細胞的修復距離均較對照組短,P均0.05。與對照組相比,sh LSINCT51#組、sh LSINCT5 3#組E-cadherin、Zo-1表達高,Vimentin表達低(P均0.05)。sh LSINCT5 1#組、sh LSINCT5 3#組在2、4 Gy劑量照射時細胞增殖能力均較對照組低(P均0.05)。結論下調(diào)人膠質(zhì)瘤細胞系U251中LSINCT5的表達,能抑制細胞增殖、轉移,并增強細胞放療敏感性。
[Abstract]:Objective to observe the effect of down-regulation of LSINCT5 on proliferation, migration and radiotherapy sensitivity of human glioma cell line U251. Methods the cell lines stably interfering with LSINCT5 (U251 sh LSINCT5 cells) were constructed by lentivirus transfer method, and the cells in sh LSINCT5 1 # group, sh LSINCT5 3 # group and GV248 transgenic control group with high interference efficiency were obtained. (MTS) assay was used to observe the proliferation of cells cultured for 0, 24, 48 and 72 h, and the cell migration ability was observed by scratch repair assay. Cell adhesion molecule (E-cadherin) was detected by, Western blotting assay. The expression of tight junction protein-1 (Zo-1) and cytoskeleton protein (Vimentin) were observed by MTS assay. The proliferation ability of the cells was observed by MTS assay after irradiation with 0, 4 and 8 Gy, respectively. Results the proliferation ability of cells in sh LSINCT5 1 # group and sh LSINCT5 3 # group was lower than that in control group at 72 h (P < 0.05). Sh LSINCT5 1 #). The repair distance of cells in sh LSINCT5 3 # group was shorter than that in control group (P 0.05). Compared with the control group, the expression of Zoo1 and Vimentin in sh LSINCT51# group and sh LSINCT5 3 # group were higher and lower than those in control group (all P 0.05). Sh LSINCT51# group, P < 0.05). The proliferation ability of sh LSINCT5 3 # group was lower than that of the control group at the dose of 2 and 4 Gy (P < 0.05), and the cell proliferation ability of the control group was lower than that of the control group (P < 0.05). Conclusion the down-regulation of LSINCT5 expression in human glioma cell line U251 can inhibit the proliferation and metastasis of human glioma cell line U251 and enhance the radiosensitivity of human glioma cell line U251.
