亞低溫對(duì)腦缺血再灌注大鼠血管內(nèi)皮生長(zhǎng)因子和低氧誘導(dǎo)因子-1α表達(dá)的影響

【摘要】:目的探討亞低溫對(duì)腦缺血再灌注大鼠內(nèi)皮生長(zhǎng)因子(VEGF)和低氧誘導(dǎo)因子-1α(HIF-1α)表達(dá)的影響。方法 30只Sprague-Dawley大鼠隨機(jī)分為假手術(shù)組、模型組和亞低溫組,每組10只。后兩組制備大腦中動(dòng)脈閉塞(MCAO)2 h再灌注動(dòng)物模型,亞低溫組進(jìn)行亞低溫處理24 h。Longa評(píng)分評(píng)估各組大鼠神經(jīng)功能;濕/干重法測(cè)定腦含水量;TTC染色觀察腦梗死體積;ELISA法檢測(cè)腦組織VEGF、HIF-1α蛋白含量;q PCR法檢測(cè)腦組織VEGF m RNA水平。結(jié)果與假手術(shù)組比,模型組神經(jīng)行為學(xué)評(píng)分升高,腦含水量及腦梗死率增加,腦組織HIF-1α蛋白、VEGF蛋白及m RNA水平上調(diào)(P0.05)。與模型組比,亞低溫組神經(jīng)神經(jīng)行為學(xué)評(píng)分顯著降低(χ~2=26.657,P0.001),腦含水量及腦梗死率降低,腦組織HIF-1α蛋白、VEGF蛋白及m RNA水平進(jìn)一步上調(diào)(P0.05)。結(jié)論亞低溫可能通過(guò)HIF-1α介導(dǎo)VEGF上調(diào),促進(jìn)缺血組織區(qū)新生血管形成,從而發(fā)揮對(duì)缺血性腦卒中的保護(hù)作用。
[Abstract]:Objective to investigate the effect of mild hypothermia on the expression of endothelial growth factor (VEGF) and hypoxia inducible factor-1 偽 (HIF-1 偽) in rats with cerebral ischemia-reperfusion. Methods 30 Sprague-Dawley rats were randomly divided into three groups: model group (n = 10), model group (n = 10) and mild hypothermia group (n = 10). The animal model of middle cerebral artery occlusion (MCAO) was established in the latter two groups. Mild hypothermia 24 h.Longa score was used to evaluate the neurological function of each group, wet / dry weight method was used to measure the brain water content, TTC staining was used to observe the volume of cerebral infarction, and mild hypothermia 24 h.Longa score was used to evaluate the neurological function of rats in each group. The content of VEGF,HIF-1 偽 protein in brain tissue was detected by ELISA method. The level of VEGF m RNA in brain tissue was detected by; q PCR method. Results compared with the pseudo-operation group, the neurobehavioral score, brain water content and cerebral infarction rate in the model group were increased, and the levels of HIF-1 偽 protein, VEGF protein and m-RNA in the brain tissue were up-regulated (P 0.05).
