【摘要】：目的探討高滲氯化鈉羥乙基淀粉40(HSH)對(duì)大鼠腦缺血-再灌注(I/R)損傷后神經(jīng)功能行為學(xué)評(píng)分,腦水腫情況,腦梗死體積以及腦組織中基質(zhì)金屬蛋白酶-9(MMP-9)和層粘連蛋白(Laminin)表達(dá)的影響。方法48只雄性SD大鼠隨機(jī)分為假手術(shù)組、模型組、實(shí)驗(yàn)1組、實(shí)驗(yàn)2組。大鼠局灶性腦I/R損傷模型制備成功后120 min,假手術(shù)組和模型組靜脈輸注0.9%生理鹽水,實(shí)驗(yàn)1組和2組分別靜脈輸注HSH 4 ml/kg和8 ml/kg。再灌注24 h后,行神經(jīng)功能行為學(xué)評(píng)分,觀察腦水腫情況,TTC染色測(cè)算腦梗死體積,用免疫印跡方法檢測(cè)缺血區(qū)腦組織MMP-9和Laminin的表達(dá)。結(jié)果與假手術(shù)組比較,模型組神經(jīng)功能行為學(xué)評(píng)分升高(P0.001);腦水腫加重;腦梗死體積增加(P0.001);MMP-9蛋白表達(dá)升高(P0.05);Laminin蛋白表達(dá)降低(P0.01)。與模型組比較,實(shí)驗(yàn)1組和2組神經(jīng)功能行為學(xué)評(píng)分降低(P0.001);腦水腫緩解;腦梗死體積減少(P0.001);實(shí)驗(yàn)1組MMP-9蛋白表達(dá)減少(P0.01),Laminin蛋白表達(dá)升高(P0.05);實(shí)驗(yàn)2組MMP-9蛋白和Laminin蛋白表達(dá)均無明顯變化。結(jié)論用HSH治療大鼠腦I/R損傷后,大鼠神經(jīng)功能行為學(xué)評(píng)分改善,腦水腫減輕,腦梗死體積減小,血腦屏障通透性降低,其機(jī)制可能與降低MMP-9和增加Laminin蛋白的表達(dá)有關(guān)。且保護(hù)作用以4 ml/kg劑量效果更佳。
[Abstract]:Objective to investigate the effects of hypertonic sodium chloride hydroxyethyl starch (40 (HSH) on neurobehavioral score and brain edema after cerebral ischemia-reperfusion (I / R) injury in rats. Effect of infarct volume and expression of matrix metalloproteinase-9 (MMP-9) and laminin (Laminin) in brain tissue. Methods 48 male SD rats were randomly divided into sham operation group, model group, experiment 1 group and experiment 2 group. HSH 4 ml/kg and 8 ml/kg. were injected intravenously into the sham operation group and model group at 120 min, after the establishment of focal cerebral I / R injury model in rats. HSH 4 ml/kg and 8 ml/kg. were injected intravenously in group 1 and group 2, respectively. 24 hours after reperfusion, neurobehavioral scores were performed to observe cerebral edema, TTC staining was used to measure the volume of cerebral infarction, and Western blot was used to detect the expression of MMP-9 and Laminin in ischemic brain tissue. Results compared with the sham-operated group, the neurobehavioral score of the model group increased (P0.001), the cerebral edema increased, the infarct volume increased (P0.001), the expression of MMP-9 protein increased (P0.05) the expression of); Laminin protein decreased (P0.01). Compared with the model group, the neurobehavioral scores of experimental group 1 and group 2 decreased (P0.001), cerebral edema alleviated, cerebral infarction volume decreased (P0.001), and cerebral infarction volume decreased (P0.001). The expression of MMP-9 protein was decreased in experiment 1 group (P0.01), Laminin protein expression was increased (P0.05), but there was no significant change in MMP-9 protein and Laminin protein expression in experiment 2 group. Conclusion after treatment with HSH, the neurobehavioral scores were improved, cerebral edema, cerebral infarction volume and blood-brain barrier permeability were decreased in rats. The mechanism may be related to the decrease of MMP-9 and the increase of Laminin protein expression. [WT5 "HZ] conclusion [WT5" BZ] [WT5 "BZ] The protective effect was better at the dose of 4 ml/kg.
