高滲氯化鈉羥乙基淀粉40對大鼠腦缺血再灌注損傷后MMP-9和層粘連蛋白表達的影響
[Abstract]:Objective to investigate the effects of hypertonic sodium chloride hydroxyethyl starch (40 (HSH) on neurobehavioral score and brain edema after cerebral ischemia-reperfusion (I / R) injury in rats. Effect of infarct volume and expression of matrix metalloproteinase-9 (MMP-9) and laminin (Laminin) in brain tissue. Methods 48 male SD rats were randomly divided into sham operation group, model group, experiment 1 group and experiment 2 group. HSH 4 ml/kg and 8 ml/kg. were injected intravenously into the sham operation group and model group at 120 min, after the establishment of focal cerebral I / R injury model in rats. HSH 4 ml/kg and 8 ml/kg. were injected intravenously in group 1 and group 2, respectively. 24 hours after reperfusion, neurobehavioral scores were performed to observe cerebral edema, TTC staining was used to measure the volume of cerebral infarction, and Western blot was used to detect the expression of MMP-9 and Laminin in ischemic brain tissue. Results compared with the sham-operated group, the neurobehavioral score of the model group increased (P0.001), the cerebral edema increased, the infarct volume increased (P0.001), the expression of MMP-9 protein increased (P0.05) the expression of); Laminin protein decreased (P0.01). Compared with the model group, the neurobehavioral scores of experimental group 1 and group 2 decreased (P0.001), cerebral edema alleviated, cerebral infarction volume decreased (P0.001), and cerebral infarction volume decreased (P0.001). The expression of MMP-9 protein was decreased in experiment 1 group (P0.01), Laminin protein expression was increased (P0.05), but there was no significant change in MMP-9 protein and Laminin protein expression in experiment 2 group. Conclusion after treatment with HSH, the neurobehavioral scores were improved, cerebral edema, cerebral infarction volume and blood-brain barrier permeability were decreased in rats. The mechanism may be related to the decrease of MMP-9 and the increase of Laminin protein expression. [WT5 "HZ] conclusion [WT5" BZ] [WT5 "BZ] The protective effect was better at the dose of 4 ml/kg.
