應(yīng)用micro-CT實(shí)現(xiàn)兔坐骨神經(jīng)顯微三維結(jié)構(gòu)可視化研究

【摘要】:目的探討通過micro-CT掃描新西蘭大白兔坐骨神經(jīng)標(biāo)本,利用三維可視化軟件Mimics17.0重建兔坐骨神經(jīng)內(nèi)部顯微三維結(jié)構(gòu)。方法取6只成年新西蘭大白兔坐骨神經(jīng)組織標(biāo)本分成A、B組(n=3),分別用1%、5%Lugol液對(duì)兩組標(biāo)本染色,于染色0.5、1.0、1.5、2.0、2.5、3.0、3.5 h時(shí),行光鏡及micro-CT觀察兩組標(biāo)本的顯像變化,將顯像良好的micro-CT圖像序列導(dǎo)入Mimics軟件,采用三維重建工具重建兔坐骨神經(jīng)神經(jīng)顯微三維結(jié)構(gòu)。結(jié)果 A組標(biāo)本在染色2.5 h、B組標(biāo)本在染色1.5 h時(shí),經(jīng)光鏡及micro-CT觀察可獲得較為清晰的顯微三維結(jié)構(gòu)圖像。圖像顯示新西蘭大白兔的坐骨神經(jīng)主要分3組神經(jīng)束,且各神經(jīng)束立體行徑相對(duì)固定,Mimics軟件測(cè)量各神經(jīng)束橫截面積分別為(0.425±0.013)、(0.038±0.007)、(0.242±0.026)mm~2,生成的數(shù)字化三維模型可在任意橫斷面觀察坐骨神經(jīng)內(nèi)部顯微結(jié)構(gòu)。結(jié)論應(yīng)用micro-CT可清晰真實(shí)顯示兔坐骨神經(jīng)顯微三維結(jié)構(gòu),為建立大樣本量周圍神經(jīng)顯微解剖學(xué)數(shù)據(jù)庫(kù)提供了可靠方法。
[Abstract]:Objective to study the reconstruction of three-dimensional microstructure of sciatic nerve in New Zealand white rabbits by micro-CT and Mimics17.0. Methods six adult New Zealand white rabbits were divided into two groups: group B (n = 3), stained with 1% and 5% Lugol solution, respectively. At the time of 0.5, 1.0, 1.5, 2.0, 2.5, 3.0, 3.5 h, respectively, the sciatic nerve tissues of 6 adult New Zealand white rabbits were stained with Lugol solution for 0.5, 1.0, 1.5, 2.0, 2.5, 3.0, 3.5 h, respectively. The imaging changes of the two groups were observed by light microscope and micro-CT. The micro-CT image sequence with good imaging was introduced into the Mimics software. The three-dimensional reconstruction tool was used to reconstruct the three-dimensional microstructure of the sciatic nerve of rabbits. Results two and a half hours after staining in group A and 1.5 hours in group B, a clear three-dimensional microstructure image could be obtained by light microscopy and micro-CT observation. The images showed that the sciatic nerve of New Zealand white rabbits was mainly divided into three groups, and the stereotactic behavior of each nerve bundle was relatively fixed. The cross-sectional area of each nerve bundle measured by Mimics software was (0.425 鹵0.013), (0.038 鹵0.007), respectively. The digital three-dimensional model produced by (0.242 鹵0.026) mm~2, can observe the microstructure of sciatic nerve on any cross-section. Conclusion micro-CT can clearly and truly display the three-dimensional structure of sciatic nerve in rabbits, which provides a reliable method for the establishment of a database of microanatomy of peripheral nerves with a large sample size.
