一鍋法合成近紅外量子點用于膠質(zhì)瘤熒光顯像與光動力治療

【摘要】:目的:合成近紅外量子點并驗證其熒光成像和光動力效應(yīng)。方法:應(yīng)用一鍋法水相合成近紅外量子點CdTe,經(jīng)表征檢測后連接RGD多肽,再用激光共聚焦顯微鏡觀察其熒光成像效果。隨后,將3~24μg/mL的CdTe-RGD分別加入U251和3T3細胞,避光培養(yǎng)24 h間,觀察量子點的細胞毒性。最后用強度為30 mW/cm~2,波長為632.8 nm的激光照射量子點處理后的細胞5~60 min,觀察其光動力效應(yīng)。結(jié)果:合成的CdTe具有良好的晶體結(jié)構(gòu)和分散性,其直徑為3.75 nm,發(fā)射波長700 nm,熒光產(chǎn)率20%。經(jīng)量子點處理后,U251細胞可熒光顯像而3T3細胞則不能。暗環(huán)境下,不同濃度CdTe-RGD處理過的U251和3T3細胞存活率都在85%以上。激光照射后,隨光照時間延長和量子點濃度的增加,U251細胞存活逐漸下降至(37±1.6)%。而3T3細胞組的存活率則無明顯變化。結(jié)論:一鍋法水相合成的CdTe-RGD具有良好的理化性質(zhì)且細胞毒性低,在生物成像和光動力治療方面具有應(yīng)用前景。
[Abstract]:Aim: to synthesize near infrared quantum dots and verify their fluorescence imaging and photodynamic effects. Methods: the near infrared quantum dots (CdTe,) were synthesized by one-pot method in aqueous phase, and then the RGD polypeptide was ligated. Then the fluorescence imaging effect was observed by confocal laser microscope (LSCM). Then, the CdTe-RGD of 3 ~ 24 渭 g / mL was added to U251 and 3T3 cells, respectively, and the cytotoxicity of QDs was observed for 24 h without light. Finally, the photodynamic effects of quantum dots treated with 30 mW/cm~2, laser at 632.8 nm were observed at 5 / 60 min,. Results: the synthesized CdTe has good crystal structure and dispersibility. Its diameter is 3.75 nm, emission wavelength 700 nm, fluorescence yield is 20%. After treatment with quantum dots, U251 cells could be visualized by fluorescence, but 3T3 cells could not. In dark environment, the survival rates of U251 and 3T3 cells treated with different concentrations of CdTe-RGD were above 85%. After laser irradiation, the survival of U251 cells decreased to (37 鹵1.6)% with the prolongation of irradiation time and the increase of quantum dot concentration. However, the survival rate of 3T3 cells did not change significantly. Conclusion: CdTe-RGD synthesized by one-pot aqueous phase has good physicochemical properties and low cytotoxicity. It has a promising future in bioimaging and photodynamic therapy.
