骨髓間充質(zhì)干細(xì)胞對(duì)實(shí)驗(yàn)性自身免疫性腦脊髓炎小鼠的Iba1、IL-1β和IL-10影響

【摘要】:目的探討骨髓間充質(zhì)干細(xì)胞(bone marrow mesenchymal stem cells,BMMSCs)對(duì)實(shí)驗(yàn)性自身免疫性腦脊髓炎(experimental autoimmune encephalomyelitis,EAE)小鼠的小膠質(zhì)細(xì)胞Iba1、IL-1β和IL-10表達(dá)的影響。方法以髓鞘少突膠質(zhì)細(xì)胞糖蛋白多肽35-55(MOG 35-55)免疫誘導(dǎo)C57BL/6雌性小鼠制備EAE小鼠模型。將30只雌性C57BL/6小鼠隨機(jī)分為3組:BMMSCs移植組、EAE組和正常組。采用免疫組化檢測(cè)腦和脊髓的Iba1(離子鈣結(jié)合銜接分子1)、IL-1β和IL-10的表達(dá)。結(jié)果 BMMSCs移植組神經(jīng)功能缺損癥狀要輕于EAE組,BMMSCs移植組腦和脊髓的Iba1和IL-1β表達(dá)水平低于EAE組(P0.05),而B(niǎo)MMSCs移植組腦和脊髓的IL-10表達(dá)水平高于EAE組(P0.05)。結(jié)論 BMMSCs能改善EAE小鼠的癥狀,其作用機(jī)制之一可能是抑制了小膠質(zhì)細(xì)胞的激活、抑制炎癥因子IL-1β及促進(jìn)抗炎因子IL-10的表達(dá)。
[Abstract]:Aim to investigate the effects of bone marrow mesenchymal stem cells (bone marrow mesenchymal stem cells,BMMSCs) on the expression of Iba1,IL-1 尾 and IL-10 in microglia of experimental autoimmune encephalomyelitis (experimental autoimmune encephalomyelitis,EAE) mice. Methods C57BL/6 female mice were immunized with myelin oligodendrocyte glycoprotein 35-55 (MOG 35-55) to establish EAE mice model. Thirty female C57BL/6 mice were randomly divided into three groups: BMMSCs transplantation group, EAE group and normal group. Immunohistochemistry was used to detect the expression of Iba1, IL-1 尾 and IL-10 in brain and spinal cord. Results the symptoms of neurological deficit in BMMSCs transplantation group were lighter than those in EAE group. The expression level of Iba1 and IL-1 尾 in brain and spinal cord of BMMSCs transplantation group was lower than that of EAE group (P0.05), while the expression level of IL-10 in brain and spinal cord of BMMSCs transplantation group was higher than that of EAE group (P0.05). Conclusion BMMSCs can improve the symptoms of EAE mice. One of its mechanisms may be that it can inhibit the activation of microglia, inhibit the expression of inflammatory factor IL-1 尾 and promote the expression of anti inflammatory factor IL-10.
