內(nèi)質(zhì)網(wǎng)應(yīng)激在帕金森病發(fā)病機(jī)制中的研究進(jìn)展

【摘要】:內(nèi)質(zhì)網(wǎng)是真核生物加工、修飾、分泌蛋白質(zhì)和儲(chǔ)存鈣離子的重要細(xì)胞器。錯(cuò)誤折疊/未折疊/突變蛋白在內(nèi)質(zhì)網(wǎng)中累積、氧化應(yīng)激和鈣離子平衡紊亂破壞了蛋白質(zhì)的清除機(jī)制,引發(fā)內(nèi)質(zhì)網(wǎng)應(yīng)激,因而激活了一種稱為未折疊蛋白反應(yīng)的適應(yīng)性應(yīng)激反應(yīng)。未折疊蛋白反應(yīng)信號(hào)由3種應(yīng)激感受分子調(diào)節(jié),它們誘導(dǎo)獨(dú)立并匯聚的信號(hào)通路來(lái)維持內(nèi)質(zhì)網(wǎng)穩(wěn)態(tài),或者在長(zhǎng)期應(yīng)激狀態(tài)下最終觸發(fā)細(xì)胞死亡。內(nèi)質(zhì)網(wǎng)應(yīng)激在帕金森病的發(fā)病進(jìn)程中有著重要作用,本綜述就內(nèi)質(zhì)網(wǎng)應(yīng)激在帕金森病中的發(fā)生、發(fā)展過(guò)程及對(duì)帕金森病的影響作一綜述。
[Abstract]:Endoplasmic reticulum (ER) is an important organelle for eukaryotic biological processing, modification, secretion of proteins and storage of calcium ions. The accumulation of misfolded / unfolded / mutant proteins in the endoplasmic reticulum (ER), oxidative stress and calcium ion balance disorder destroyed the mechanism of protein clearance and induced endoplasmic reticulum stress. This activates an adaptive stress response called the unfolded protein response. Unfolded protein response signals are regulated by three stress-sensing molecules, which induce independent and convergent signaling pathways to maintain endoplasmic reticulum homeostasis or eventually trigger cell death under long-term stress. Endoplasmic reticulum stress plays an important role in the pathogenesis of Parkinson's disease. This review reviews the occurrence and development of endoplasmic reticulum stress in Parkinson's disease and its effects on Parkinson's disease.
