頸交感神經(jīng)阻滯對(duì)糖尿病大鼠腦線粒體功能的影響

【摘要】:目的觀察頸交感神經(jīng)阻滯對(duì)糖尿病大鼠腦線粒體功能的影響。方法雄性成年大鼠40只隨機(jī)分為3組:對(duì)照組(n=10,腹腔注射注射等量緩沖液),糖尿病組(n=15,采用STZ糖尿病模型按65 mg/kg劑量腹腔1次注射1%STZ制備糖尿病大鼠),糖尿病+頸交感神經(jīng)阻滯組(n=15,按65 mg/kg劑量腹腔1次注射1%STZ,糖尿病成模后8周給予頸交感神經(jīng)阻滯治療)。造模后12周水迷宮實(shí)驗(yàn)觀察各組大鼠的認(rèn)知功能。并于12周處死大鼠,采用Clark氧電極法測(cè)定大鼠海馬線粒體的呼吸功能,免疫組化檢測(cè)海馬IL-6、TNF-α的表達(dá)。結(jié)果造模后12周,糖尿病組存活12只,阻滯組存活13只。糖尿病組、阻滯組與對(duì)照組相比,逃避潛伏期明顯延長、穿越平臺(tái)的次數(shù)明顯減少(P0.05)。糖尿病組線粒體呼吸功能為R3(28.51±3.01)、R4(13.93±1.16)、RCR(2.09±0.27),阻滯組的為R3(45.26±4.21)、R4(11.99±1.11)、RCR(3.52±0.32),對(duì)照組為R3(66.31±5.65)、R4(12.97±3.38)、RCR(4.78±0.68),糖尿病組和阻滯組線粒體呼吸功能均較對(duì)照組組明顯下降,主要表現(xiàn)為R3和RCR值顯著降低(P0.05);但阻滯組的R3和RCR高于糖尿病組(P0.05)。在海馬CA1區(qū),糖尿病組IL-6、TNF-α表達(dá)為(53.32±6.29)、(63.21±13.24),阻滯組為(28.14±3.82)、(32.76±5.98),均高于對(duì)照組(13.87±2.54)、(11.09±8.87)(P0.05)。阻滯組明顯低于糖尿病組,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論頸交感神經(jīng)阻滯可改善糖尿病大鼠腦線粒體功能,其機(jī)制可能與其下調(diào)IL-6、TNF-α有關(guān)。
[Abstract]:Objective to observe the effect of cervical sympathetic block on mitochondrial function in diabetic rats. Methods Forty adult male rats were randomly divided into three groups: control group (n = 10, intraperitoneal injection of the same amount of buffer), diabetic group (n = 15, STZ diabetes model was injected intraperitoneally with 1%STZ at a dose of 65 mg/kg), diabetes mellitus (DM). Cervical sympathetic nerve block group (NN15, 65 mg/kg intraperitoneal injection of 1 STZ, diabetes model 8 weeks after the treatment of cervical sympathetic nerve block). The cognitive function of rats in each group was observed by water maze experiment 12 weeks after modeling. The respiratory function of hippocampus mitochondria was measured by Clark oxygen electrode method and the expression of IL-6,TNF- 偽 in hippocampus was detected by immunohistochemistry. Results 12 weeks after modeling, 12 rats survived in diabetic group and 13 survived in block group. Compared with the control group, the escape latency was significantly prolonged and the frequency of crossing the platform was significantly decreased in the diabetic group and the block group (P0.05). R3 (28.51 鹵3.01) R4 (13.93 鹵1.16) RCR (2.09 鹵0.27), R3 (45.26 鹵4.21) R4 (11.99 鹵1.11) RCR (3.52 鹵0.32), R3 (66.31 鹵5.65) R4 (12.97 鹵3.38) RCR and block group (4.78 鹵0.68) in diabetic group and block group respectively. R3 and RCR decreased significantly (P0.05), but R3 and RCR in block group were higher than that in diabetic group (P0.05). In hippocampal CA1 area, the expression of IL-6,TNF- 偽 in diabetic group (53.32 鹵6.29), (63.21 鹵13.24) and block group (28.14 鹵3.82), (32.76 鹵5.98) was higher than that in control group (13.87 鹵2.54), (11.09 鹵8.87) (P0.05). Block group was significantly lower than diabetes group, the difference was statistically significant (P0.05). Conclusion Cervical sympathetic nerve block can improve mitochondrial function in diabetic rats, and its mechanism may be related to its down-regulation of IL-6,TNF- 偽.
