脾和腰部淋巴結(jié)切除緩解大鼠脛神經(jīng)損傷后疼痛慢性化

【摘要】:目的探討外周淋巴器官在成年大鼠脛神經(jīng)永久性橫斷傷(即改良型備用性神經(jīng)損傷,modified spared nerve injury,mSNI)后機械性疼痛異常的慢性轉(zhuǎn)化中的作用。方法 30只成年雄性Sprague-Dawley(SD)大鼠隨機分為3組,每組10只動物,分別用于脾臟切除、腰部淋巴結(jié)切除和右側(cè)乆窩淋巴結(jié)-腹股溝淋巴結(jié)聯(lián)合切除實驗。每項實驗的10只動物再隨機分為外周淋巴器官切除手術(shù)組(n=5)和假手術(shù)組(n=5)。所有動物根據(jù)實驗分組進行相應(yīng)的外周淋巴器官切除的手術(shù)或者假手術(shù),術(shù)后1周進行右側(cè)mSNI手術(shù)。所有動物于外周淋巴器官切除術(shù)前、右側(cè)mSNI手術(shù)前、mSNI手術(shù)后7d和mSNI手術(shù)后14d進行von Frey纖維試驗,測試mSNI手術(shù)同側(cè)和對側(cè)大鼠后爪足底面腓腸神經(jīng)和隱神經(jīng)皮膚區(qū)域特異性的疼痛反應(yīng)。結(jié)果脾臟和腰部淋巴結(jié)切除明顯緩解mSNI后7d和(或)14d的大鼠手術(shù)側(cè)后爪足底面腓腸神經(jīng)和隱神經(jīng)皮膚區(qū)域的機械性疼痛異常。然而,右側(cè)胭窩淋巴結(jié)-腹股溝淋巴結(jié)聯(lián)合切除對mSNI后7d和14d的大鼠手術(shù)側(cè)后爪足底面腓腸神經(jīng)和隱神經(jīng)皮膚區(qū)域的機堿性疼痛異常沒有顯著影響。結(jié)論脾臟和腰部淋巴結(jié)參與成年大鼠mSNI后機械性疼痛異常的慢性轉(zhuǎn)化。
[Abstract]:Objective to investigate the role of peripheral lymphoid organs in the chronic transformation of abnormal mechanical pain after permanent transection of tibial nerve (modified spared nerve injury,mSNI) in adult rats. Methods Thirty adult male Sprague-Dawley (SD) rats were randomly divided into 3 groups, 10 animals in each group, which were used for splenectomy, lumbar lymphadenectomy, and combined excision of right and right ventral lymph nodes and inguinal lymph nodes. Ten animals in each experiment were randomly divided into two groups: peripheral lymphotomy group (N5) and sham operation group (N5). All animals were treated with peripheral lymphotomy or sham-operation according to the experimental group, and the right mSNI was performed 1 week after operation. All animals were tested for von Frey fibers before peripheral lymphotomy, 7 days after right mSNI operation and 14 days after mSNI operation. The region specific pain response of sural nerve and saphenous nerve was measured in ipsilateral and contralateral mSNI rats. Results splenectomy and lumbar lymphadenectomy significantly alleviated the abnormal mechanical pain in the sural nerve and saphenous nerve region of the lateral sural nerve and saphenous nerve of the rats at 7 and / or 14 days after mSNI. However, the combined excision of the right popliteal and inguinal lymph nodes had no significant effect on the pathogenesis of alkaline pain in the sural nerve and saphenous nerve skin region of the lateral paw of rats 7 and 14 days after mSNI. Conclusion spleen and lumbar lymph nodes participate in the chronic transformation of abnormal mechanical pain after mSNI in adult rats.
