14-3-3β蛋白對膠質(zhì)瘤細(xì)胞生物學(xué)行為的影響

【摘要】:目的探討14-3-3β蛋白對膠質(zhì)瘤細(xì)胞系生物學(xué)行為的影響。方法應(yīng)用分子生物學(xué)技術(shù)構(gòu)建14-3-3β蛋白的真核表達(dá)質(zhì)粒pc DNA 3.1(-)-14-3-3β及干擾質(zhì)粒pc DNATM6.2-GW/Em GFPmiR-14-3-3β,應(yīng)用MTT、集落形成、Transwell侵襲試驗研究14-3-3β蛋白對人膠質(zhì)瘤細(xì)胞株U251增殖及侵襲等生物學(xué)行為的影響。結(jié)果過表達(dá)14-3-3β能夠明顯促進U251細(xì)胞(人膠質(zhì)瘤系)的侵襲和增殖過程;而應(yīng)用干擾質(zhì)粒抑制14-3-3β表達(dá)后,U251細(xì)胞的侵襲和增殖能力則受到顯著抑制。結(jié)論 14-3-3β對于U251細(xì)胞(人膠質(zhì)瘤系)蛋白的侵襲和增殖能力起到明確的促進作用,其在人類膠質(zhì)瘤的發(fā)生和發(fā)展過程中可能也起到促癌作用。
[Abstract]:Objective to investigate the effect of 14-3-3 尾 protein on the biological behavior of glioma cell line. Methods the eukaryotic expression plasmid pc DNA 3.1 (-) -14-3-3 尾 and the interfering plasmid pc DNATM6.2-GW/Em GFPmiR-14-3-3 尾 of 14-3-3 尾 protein were constructed by molecular biology technique. The effects of 14-3-3 尾 protein on the proliferation and invasion of human glioma cell line U251 were studied by MTT, colony forming transwell invasion test. Results overexpression of 14-3-3 尾 could significantly promote the invasion and proliferation of U251 cells (human glioma cells), while the ability of invasion and proliferation of U251 cells was significantly inhibited by using interference plasmids to inhibit the expression of 14-3-3 尾. Conclusion 14-3-3 尾 can promote the invasion and proliferation of U251 cell line (human glioma cell line), and it may also play a role in the carcinogenesis and development of human glioma.
