腺苷A2A受體對新生小鼠離體延髓腦片基本節(jié)律性呼吸放電的調(diào)節(jié)作用

【摘要】:目的:探討腺苷A2A受體對新生小鼠離體延髓腦片基本節(jié)律性呼吸放電(RRDA)的調(diào)節(jié)作用。方法:制備新生小鼠離體延髓腦片,分別使用含0.0、2.5、5.0、10.0、20.0 nmol/L腺苷A2A受體激動劑CGS21680或拮抗劑SCH58261的人工腦脊液(ACSF)灌流腦片,每個濃度灌流10 min,在每個濃度灌流第6 min時,使用吸附電極記錄腦片RRDA,分析吸氣時程(TI)、放電積分幅度(IA)和呼吸周期(RC)。結(jié)果:CGS21680可以縮短RC,增加TI和IA(P0.001),對RRDA有興奮作用。SCH58261則可以延長RC,減小TI和IA(P0.001),對RRDA有抑制作用。結(jié)論:腺苷A2A受體參與調(diào)節(jié)新生小鼠延髓RRDA,激活腺苷A2A受體對RRDA有興奮作用。
[Abstract]:Aim: to investigate the regulatory effect of adenosine A 2A receptor on the basic rhythmic respiratory discharge (RRDA) in isolated medulla oblongata of newborn mice. Methods: in vitro medulla oblongata slices of newborn mice were prepared and perfused with the artificial cerebrospinal fluid (ACSF) containing 0.02.55.010.0 nmol/L A2A receptor agonist CGS21680 or antagonist SCH58261 for 10 minutes, at 6 min in each concentration. The absorption electrode was used to record the RRDAs of brain slices. The integral amplitude (IA) of inspiratory (TI), discharge and the (RC). Of respiratory cycle were analyzed. Results: CGS21680 could shorten RCand increase TI and IA (P0.001), but it could prolong RRDA, decrease TI and IA (P0.001) and inhibit RRDA. Conclusion: adenosine A 2A receptor is involved in the regulation of medulla oblongata of newborn mice, and the activation of adenosine A 2A receptor can stimulate RRDA.
