TACI-Ig融合蛋白對(duì)小鼠視神經(jīng)病理?yè)p傷的影響

【摘要】:目的·探討腹腔注射TACI-Ig融合蛋白對(duì)小鼠視神經(jīng)炎癥浸潤(rùn)、髓鞘完整性等的影響。方法·將24只C57BL/6J小鼠隨機(jī)分為4組,分別為空白對(duì)照組、生理鹽水注射組、TACI-Ig低劑量注射組(0.4 mg/kg)、TACI-Ig高劑量注射組(4 mg/kg),每組6只。TACI-Ig隔日腹腔注射1次,生理鹽水對(duì)照組按照同樣方式注射生理鹽水0.2 mL,空白對(duì)照組不給予任何干預(yù)。20日后將小鼠麻醉灌注,取眼球和視神經(jīng)組織,石蠟包埋,切片后行蘇木精-伊紅(H-E)染色和固藍(lán)(LFB)染色,分別觀察炎癥細(xì)胞浸潤(rùn)情況和髓鞘脫失情況。結(jié)果·H-E染色可見(jiàn)空白對(duì)照組和生理鹽水對(duì)照組著色均勻,視神經(jīng)纖維排列緊密、規(guī)則;TACI-Ig低劑量注射組(0.4 mg/kg)視神經(jīng)結(jié)構(gòu)與兩對(duì)照組類似;高劑量注射組(4 mg/kg)細(xì)胞均勻分布,可觀察到炎癥細(xì)胞少量浸潤(rùn),炎癥細(xì)胞浸潤(rùn)評(píng)分與其他組比較無(wú)明顯差異(P=0.610 3)。所有組別視網(wǎng)膜結(jié)構(gòu)清晰、層次分明,神經(jīng)節(jié)細(xì)胞單層緊密排列、胞體完整、胞核可見(jiàn)、分布均勻,各組實(shí)驗(yàn)動(dòng)物視網(wǎng)膜各層結(jié)構(gòu)相比較并無(wú)明顯差異。經(jīng)LFB染色后光學(xué)顯微鏡下觀察,發(fā)現(xiàn)視神經(jīng)髓鞘均無(wú)明顯脫失和損傷情況,4組之間比較差異無(wú)統(tǒng)計(jì)學(xué)意義(P=0.473 6)。結(jié)論·低劑量(0.4 mg/kg)TACI-Ig腹腔注射對(duì)視神經(jīng)、視網(wǎng)膜神經(jīng)節(jié)細(xì)胞無(wú)損傷,高劑量(4 mg/kg)可能引起視網(wǎng)膜區(qū)域炎癥少量浸潤(rùn)。
[Abstract]:Objective to investigate the effects of intraperitoneal injection of TACI-Ig fusion protein on inflammatory infiltration and myelin integrity of optic nerve in mice. Methods Twenty-four C57BL / 6J mice were randomly divided into 4 groups: control group, saline injection group (0.4 mg/kg), TACI-Ig high dose injection group (4 mg/kg), 6 mice in each group were intraperitoneally injected with TACI-Ig every other day. The control group was injected with 0.2 mL normal saline in the same way, and the blank control group was not given any intervention. After 20 days, the mice were anesthetized and perfused, the eyeballs and optic nerve tissue were taken and embedded in paraffin. Hematoxylin-eosin (H-E) staining and solid blue (LFB) staining were used to observe the infiltration of inflammatory cells and demyelination of myelin. Results H-E staining showed that the optic nerve fibers in the blank control group and the saline control group were uniform, the optic nerve fibers were arranged closely, the structure of optic nerve in the regular TACI-Ig low-dose injection group (0.4 mg/kg) was similar to that in the two control groups, and the cells in the high-dose injection group (4 mg/kg) were evenly distributed. A small amount of inflammatory cell infiltration was observed, and the score of inflammatory cell infiltration was not significantly different from that of other groups (P0. 6103). In all groups, the retinal structure was clear and stratified, the ganglion cells were closely arranged in a single layer, the cell body was intact, the nucleus was visible and distributed evenly, and there was no significant difference in the structure of each layer of retina among the experimental animals. Under the optical microscope after LFB staining, there was no significant difference in demyelination and injury of optic nerve between the four groups (P0. 4736). Conclusion low dose (0.4 mg/kg) TACI-Ig intraperitoneal injection can not damage the optic nerve and retinal ganglion cells, and high dose (4 mg/kg) may cause a small amount of inflammation infiltration in the retina region.
