羥基紅花黃素A對(duì)腦缺血再灌注后缺血半暗帶自噬活性的調(diào)節(jié)作用

【摘要】:目的:探討腦缺血再灌注后羥基紅花黃素A(hydroxysafflor yellow A,HYSA)對(duì)缺血半暗帶自噬活性的調(diào)控機(jī)制。方法:構(gòu)建雄性SD大鼠腦缺血90 min再灌注模型,分為假手術(shù)組、腦缺血再灌注(cerebral ischmia/reperfusion,I/R)組、I/R+HSYA組和假手術(shù)+HSYA組,分別在再灌注后6 h、1 d、3 d和7 d對(duì)大鼠進(jìn)行改良神經(jīng)功能缺損評(píng)分(modified neurological severity score,m NSS),同時(shí)檢測(cè)缺血半暗帶的自噬活性、凋亡及干擾素β(IFN-β)表達(dá)。結(jié)果:與假手術(shù)組比較,I/R組缺血半暗帶在6 h~7 d內(nèi)可見(jiàn)LC3-Ⅱ蛋白表達(dá)增高及SQSTM1/P62蛋白降解,提示自噬激活;與I/R組比較,I/R+HSYA組的自噬活性在1 d和3 d時(shí)明顯升高(P0.05),7 d時(shí)則明顯降低(P0.05)。同時(shí),I/R組的IFN-β表達(dá)較假手術(shù)組在6 h時(shí)升高(P0.05),1 d和3 d時(shí)降低(P0.05),7 d時(shí)恢復(fù)正常;而I/R+HSYA組的IFN-β表達(dá)在1 d和3 d時(shí)明顯高于假手術(shù)組和I/R組(P0.05),并且3 d時(shí)的細(xì)胞凋亡少于I/R組,m NSS評(píng)分自4 d起明顯低于I/R組(P0.05)。結(jié)論:HSYA可動(dòng)態(tài)調(diào)節(jié)缺血半暗帶的自噬活性和IFN-β表達(dá),從而改善腦缺血再灌注損傷。
[Abstract]:Aim: to investigate the regulatory mechanism of hydroxysafflor A (hydroxysafflor yellow HYSA on autophagy activity in ischemic penumbra after cerebral ischemia reperfusion. Methods: male Sprague-Dawley rats were divided into sham operation group, cerebral ischemia reperfusion I / R group and sham operation HSYA group.
