三叉神經(jīng)痛大鼠三叉神經(jīng)根區(qū)Fyn的表達(dá)變化

【摘要】:目的:在三叉神經(jīng)痛(trigeminal neuralgia,TN)動(dòng)物模型基礎(chǔ)上,研究三叉神經(jīng)根區(qū)(trigeminal nerve root entry zone,TREZ)Fyn蛋白的表達(dá)。方法:通過(guò)三叉神經(jīng)根慢性壓迫損傷誘導(dǎo)建立大鼠TN動(dòng)物模型,von Frey絲檢測(cè)大鼠口面部術(shù)前1 d和術(shù)后1、3、7、14、21 d和28 d的機(jī)械性痛敏,然后在大鼠分別存活7、14、21 d和28 d時(shí)灌注取材,采用免疫熒光染色方法以及蛋白質(zhì)印跡(Western Blot)技術(shù)檢測(cè)三叉神經(jīng)根區(qū)Fyn的表達(dá)變化。結(jié)果:(1)行為學(xué)結(jié)果顯示:TN模型組大鼠術(shù)后14 d的機(jī)械性痛敏閾值明顯降低,與假手術(shù)(Sham)組相比有顯著性差異(P0.05);(2)免疫熒光染色顯示:TN模型組大鼠術(shù)后三叉神經(jīng)根區(qū)Fyn陽(yáng)性細(xì)胞增多,Sham組未見(jiàn)明顯變化。(3)Western Blot顯示:TN模型組大鼠術(shù)后7 d Fyn表達(dá)量相對(duì)Sham組降低(P0.05),術(shù)后14 d降至最低;術(shù)后21 d Fyn表達(dá)上調(diào),但仍低于Sham組;術(shù)后28 d TN組Fyn表達(dá)量明顯上升且高于Sham組。結(jié)論:大鼠TREZ慢性壓迫損傷后,Fyn表達(dá)在TN早期降低,而在TN后期Fyn表達(dá)升高,提示Fyn可能參與TN動(dòng)物模型的發(fā)病過(guò)程。
[Abstract]:Aim: to study the expression of Fyn protein in the trigeminal root region of trigeminal neuralgia (trigeminal neuralgia TN). Methods: the rat TN animal model was established by chronic compression injury of trigeminal nerve root. Von Frey wire was used to detect the mechanical pain sensitivity of rats at 1 day before operation and 1 day after operation, and 21 days and 28 days after operation, and then the rats were perfused at 714 days and 28 days, respectively. The expression of Fyn in trigeminal nerve root was detected by immunofluorescence staining and Western blot. Results: (1) Behavioral results showed that the mechanical pain sensitivity threshold of the rats in the weight TN group was significantly lower than that in the control group on the 14th day after operation. There was significant difference between sham group and sham group (P0.05); (2) immunofluorescence staining showed that Fyn positive cells in the trigeminal nerve root area of the rats in the group of 10 TN were increased after operation. (3) Western blot showed that the expression of Fyn was not changed 7 days after operation in the control group. (3) the results of Western blot showed that the expression of Fyn was not changed in the control group on the 7th day after operation. Compared with Sham group, it was decreased (P0.05), and decreased to the lowest on the 14th day after operation. The expression of Fyn was up-regulated 21 days after operation, but still lower than that of Sham group, and the expression of Fyn in TN group was significantly higher than that in Sham group on the 28th day after operation. Conclusion: the expression of Fyn decreased in the early stage of TN, but increased in the late stage of TN, suggesting that Fyn may be involved in the pathogenesis of TN animal model.
