長鏈非編碼RNA在腦膠質(zhì)瘤中差異表達的相關(guān)研究

【摘要】:神經(jīng)膠質(zhì)瘤是腦神經(jīng)神經(jīng)系統(tǒng)是最為常見的惡性腫瘤,致死率高、復(fù)發(fā)率高、死亡率高,,目前對于膠質(zhì)瘤的治療方法包括手術(shù)治療、化學(xué)藥物治療、放射治療、新生物治療等,然而預(yù)后的改善仍然較為有有限。膠質(zhì)瘤治療困難的主要原因包括:腫瘤細胞通常過度的增殖和廣泛的組織浸潤、發(fā)病位置難以進行手術(shù)切除、病理類型不易確定等。因此尋找有效的神經(jīng)膠質(zhì)瘤分子診斷標記和分子靶點,阻斷腫瘤細胞的惡性增殖,提升治療效率一直是研究的熱點。長鏈非編碼RNA是人類轉(zhuǎn)錄組中一大類重要的調(diào)控分子,是指那些長度大于200nt的非編碼RNA,其序列上缺乏明顯的有開放讀碼框,不能編碼蛋白,可以在轉(zhuǎn)錄前、轉(zhuǎn)錄后和表觀遺傳學(xué)多個水平參與對靶基因的調(diào)控而發(fā)揮重要的生物學(xué)功能。已有研究證明lncRNA幾乎涉及了生物體所有生理和病理過程,lncRNA的異常表達與臨床上許多腫瘤及非腫瘤疾病關(guān)系密切,因此被寄望成為為腫瘤治療的重要分子靶點,然而目前我們對于lncRNA在膠質(zhì)瘤發(fā)生發(fā)展過程中的重要作用,仍知之甚少。 本論文開展lncRNA的異常表達與腦膠質(zhì)瘤的關(guān)聯(lián)研究,具體選取了3種在其他實體腫瘤中被發(fā)現(xiàn)有重要功能的3種lncRNA,結(jié)腸癌差異表達(CRNDE)、尿路上皮癌抗原(UCA1)、肺腺癌轉(zhuǎn)移相關(guān)轉(zhuǎn)錄因子1(MALAT1),利用熒光定量PCR檢測,3種lncRNA在膠質(zhì)瘤組織和癌旁組織中的表達差異,并將lncRNA的表達量變化情況與膠質(zhì)瘤的病理分級和復(fù)發(fā)情況相關(guān)聯(lián),開展臨床治療。同時我們還在嘗試在膠質(zhì)瘤細胞中外源性過表達lncRNAUCA1,觀察其對于腫瘤細胞增殖能力的影響,并探討可能的作用機制。
[Abstract]:Glioma is the most common malignant tumor in the cerebral nervous system, with high mortality, high recurrence rate and high mortality. At present, the treatment of glioma includes surgical treatment, chemotherapy, radiotherapy, new biological therapy, etc. However, the improvement of prognosis is still limited. The main reasons for the difficulty in the treatment of glioma include: excessive proliferation of tumor cells, extensive tissue infiltration, difficulty in surgical excision and difficult identification of pathological types, etc. Therefore, finding effective molecular diagnostic markers and molecular targets for glioma, blocking the malignant proliferation of tumor cells and improving the therapeutic efficiency has been a hot topic. Long strand noncoding RNAs are important regulatory molecules in human transcriptome. They are those that are longer than 200nt. They lack an open reading frame and can not encode proteins, so they can be transcripted before transcription. Posttranscriptional and epigenetic levels play important biological roles in the regulation of target genes. Studies have shown that the abnormal expression of lncRNA involved in almost all biological physiological and pathological processes is closely related to many tumor and non-tumor diseases in clinic, so it is expected to be an important molecular target for tumor therapy. However, we still know little about the important role of lncRNA in the development of glioma. Three kinds of LNcRNAs which were found to have important functions in other solid tumors were selected. The differential expression of CRNDEG in colon cancer, UCA1 antigen in urothelial epithelium, and transcriptional factor 1MALAT1 associated with metastasis of lung adenocarcinoma were selected. Three LNcRNAs in glia were detected by fluorescence quantitative PCR. The difference of expression between tumor tissues and paracancerous tissues, The expression of lncRNA was correlated with the pathological grade and recurrence of glioma, and the clinical treatment was carried out. At the same time, we are trying to overexpression lncRNAUCA1 in glioma cells, to observe the effect of UCA1 on the proliferation of tumor cells, and to explore the possible mechanism.
