石杉堿甲對帕金森病模型大鼠乙酰膽堿酯酶及ATP酶活性的影響

【摘要】:目的探討石杉堿甲對帕金森模型病大鼠乙酰膽堿酯酶(AchE)及ATP酶活性的影響。方法選擇45只雄性SD大鼠為研究對象,采用隨機數(shù)字表法分為空白對照組、模型組、石杉堿甲組各15只。模型組、石杉堿甲組采用6-羥基多巴胺腦立體定向單側(cè)紋狀體雙靶點微量注射法制備帕金森大鼠模型,石杉堿甲組給予0.1mg·kg-1·d-1石杉堿甲灌胃,空白對照組和模型組灌入等容量蒸餾水。比較3組大鼠空間記憶能力、紋狀體多巴胺(DA)水平及AchE水平、腦內(nèi)黑質(zhì)Ca~(2+)-ATPase、Na~+-K~+-ATPase、Ca~(2+)-Mg~(2+)-ATPase活性。結(jié)果模型組信息游泳時間、選擇游泳時間明顯長于空白對照組(t=6.907、6.966,P0.05),石杉堿甲組大鼠信息游泳時間、選擇游泳時間明顯短于模型組(t=4.572、3.444,P0.05),且石杉堿甲組大鼠且明顯短于模型組(t=2.421、3.912,P0.05);模型組大鼠腦組織多巴胺明顯低于空白對照組,AchE明顯高于空白對照組(t=12.517,6.834,P0.05),石杉堿甲組大鼠腦組織多巴胺明顯高于模型組,AchE明顯低于模型組(t=6.388、4.719,P0.05),且石杉堿組大鼠腦組織多巴胺明顯低于空白對照組,AchE明顯高于空白對照組(t=6.055、2.024,P0.05);模型組大鼠腦黑質(zhì)中Ca~(2+)-ATPase、Na~+-K~+-ATPase、Ca~(2+)-Mg~(2+)-ATPase水平均明顯低于空白對照組(t=20.326、15.2225、12.517,P0.05),石杉堿甲組大鼠腦黑質(zhì)中Ca~(2+)-ATPase、Na~+-K~+-ATPase、Ca~(2+)-Mg~(2+)-ATPase水平均高于模型組(t=16.771、9.516、7.480,P0.05),且大鼠腦黑質(zhì)中Ca~(2+)-ATPase、Na~+-K~+-ATPase、Ca~(2+)-Mg~(2+)-ATPase水平均低于模型組(t=3.901、5.844、5.848,P0.05)。結(jié)論石杉堿甲有助于提高帕金森大鼠空間記憶能力,可能與抑制AchE表達、提高腦組織中多巴胺及ATP酶的活性有關(guān)。
[Abstract]:Objective to investigate the effects of Huperzine A on the activities of acetylcholinesterase (ache) and ATP in Parkinson's disease (PD) rats. Methods Forty-five male Sprague-Dawley rats were randomly divided into control group (n = 15) and Huperzine A group (n = 15). Model group, Huperzine A group, 6-hydroxydopamine stereotactic unilateral striatal microinjection was used to make Parkinson rat model. Huperzine A group was given 0.1mg kg-1 d-1 Huperzine A intragastric perfusion. Blank control group and model group were perfused with distilled water of equal capacity. The spatial memory ability, striatal dopamine (DA) level and AchE level in the three groups were compared, and the activity of Ca~(2 ~ -K ~ -ATPasesi-Na ~ -K ~ -AT Pasea-CaO2-Mg-PS-ATPase in the substantia nigra was compared. Results the information swimming time in the model group was significantly longer than that in the blank control group. The swimming time in the model group was significantly shorter than that in the model group, and that in the Huperzine A group was significantly shorter than that in the model group, and the dopamine in the brain tissue of the model group was significantly lower than that in the blank control group, and that in the model group was significantly higher than that in the blank control group (t12.5176.834, P 0.05), and that in the Huperzine A group, the brain tissue of the model group was significantly lower than that of the blank control group, and that of the rats in the Huperzine A group was significantly higher than that in the control group. The tissue dopamine was significantly higher than that in the model group (6.3884.719 P0.05), and the dopamine in the brain tissue of the Huperzine group was significantly lower than that of the blank control group (P 0.05). The contents of Ca~(2 -ATPaseNa ~ -K ~ -ATPase2 + -ATPase water in the substantia nigra of the model group were significantly higher than that of the blank control group (6.055 ~ 2.024) P0.05; the content of Ca~(2 -ATPaseNa ~ -K ~ -ATPase2 in the substantia nigra of the model group was significantly higher than that of the blank control group (P0.05). The average level of Ca~(2 in the substantia nigra of rats in Huperzine A group was significantly lower than that in the blank control group. The levels of Ca~(2 ~ -K ~ -ATPaseCaO2 + -MgP0. 05-ATPase in the substantia nigra of rats were all higher than those in the model group (t16.7719.5167.480P0.05), and the levels of Ca~(2 -ATPaseNa ~ -K ~ -ATPase2 -Mg2 in the substantia nigra of rats were lower than those of the model group. Conclusion Huperzine A can improve the spatial memory ability of Parkinson's rats, which may be related to the inhibition of AchE expression and the increase of dopamine and ATP enzyme activities in brain tissue.
