【摘要】:目的利用小鼠誘導(dǎo)性多能干細(xì)胞(iPSCs)建立大腦皮質(zhì)類器官并分析其生物學(xué)特性。方法首先將生長(zhǎng)在飼養(yǎng)層細(xì)胞上的iPSCs克隆消化成單細(xì)胞,懸浮培養(yǎng)使其自發(fā)形成擬胚體。隨后,對(duì)其進(jìn)行神經(jīng)上皮分化的誘導(dǎo)。在合適時(shí)間將其嵌入Matrigel基質(zhì)膠做的懸滴中,搖動(dòng)培養(yǎng),形成大腦皮質(zhì)類器官;最后利用免疫熒光、Dio示蹤、透射電子顯微鏡等技術(shù)檢測(cè)大腦皮質(zhì)類器官的生物學(xué)特性。結(jié)果大腦皮質(zhì)類器官有神經(jīng)上皮、神經(jīng)元及神經(jīng)膠質(zhì)細(xì)胞的分化特征;具有簡(jiǎn)單的片層化結(jié)構(gòu),如皮質(zhì)板和分子層,且大腦皮質(zhì)類器官具有神經(jīng)再生及修復(fù)能力。結(jié)論利用iPSCs成功建立了大腦皮質(zhì)類器官模型,且該模型具有與活體大腦皮質(zhì)相似的特征,且具有神經(jīng)再生能力。
[Abstract]:Objective to establish cerebral cortex organs and analyze their biological characteristics by using induced pluripotent stem cells (iPSCs) in mice. Methods iPSCs grown in feeder layer cells was cloned and digested into single cells, which were cultured in suspension culture to form embryoid body spontaneously. Then, the differentiation of neuroepithelium was induced. At the appropriate time, it was embedded in the suspension of Matrigel gel to shake and culture to form cerebral cortical organs. Finally, the biological characteristics of cerebral cortical organs were detected by immunofluorescence tracing and transmission electron microscopy. Results the cerebral cortical organs had the differentiation characteristics of neuroepithelium, neurons and glial cells, and had simple lamellar structures, such as cortical plate and molecular layer, and the cerebral cortical organs had the ability of nerve regeneration and repair. Conclusion the model of cerebral cortical organs was successfully established by using iPSCs. The model has the characteristics similar to that of the in vivo cerebral cortex and has the ability of nerve regeneration.
