雷帕霉素對PC12細(xì)胞氧糖剝奪損傷的保護作用

【摘要】:目的探討雷帕霉素對PC12細(xì)胞氧糖剝奪損傷(oxygen and glucose deprivation,OGD)的作用。方法選取處于對數(shù)生長期的PC12細(xì)胞,分為正常組、OGD組、溶劑組和1 nmol、10 nmol、100 nmol、500 nmol雷帕霉素組,OGD同時給予雷帕霉素處理,8 h后通過倒置顯微鏡觀察細(xì)胞形態(tài)學(xué)的變化、MTT法檢測細(xì)胞存活率的變化以及LDH活性檢測細(xì)胞受損程度。結(jié)果與正常組相比,單純OGD組細(xì)胞活性明顯下降,且具有統(tǒng)計學(xué)意義(P0.05);與溶劑組相比,1、10、100、500 nmol雷帕霉素組細(xì)胞存活率均存在不同程度的增高,其中10、100、500 nmol組具有統(tǒng)計學(xué)意義(P0.05)。結(jié)論雷帕霉素對PC12細(xì)胞OGD損傷具有保護作用。
[Abstract]:Objective to investigate the effect of rapamycin on oxygen and glucose deprivation (OGD) in PC12 cells. Methods PC12 cells in logarithmic growth phase were divided into normal group and control group. The cells were treated with rapamycin for 8 hours in solvent group and 10nmol / 100nmol / 500 nmol rapamycin group. The changes of cell morphology were observed by inverted microscope. The changes of cell survival rate and the degree of cell damage were detected by LDH activity. Results compared with the normal group, the cell activity of the OGD group was significantly decreased, and the cell survival rate of the 10100500 nmol group was significantly higher than that of the solvent group (P 0.05), and the survival rate of the 10100500 nmol group was significantly higher than that of the solvent group (P 0.05). Conclusion Rapamycin has protective effect on OGD damage of PC12 cells.
