miRNA-21、miRNA-221、miRNA-222在膠質(zhì)瘤病人血液中的表達(dá)及其意義

【摘要】:目的:檢測(cè)miRNA-21、miRNA-221、miRNA-222在腦膠質(zhì)瘤病人血液中的表達(dá)和探討miRNA-21、miRNA-221、miRNA-222與腦膠質(zhì)瘤發(fā)生發(fā)展的相關(guān)性,為腦膠質(zhì)瘤的早期診斷和進(jìn)展判斷提供了可行的方法和途徑。 方法:收集60例術(shù)后病理證實(shí)為膠質(zhì)瘤患者的血液標(biāo)本作為實(shí)驗(yàn)組和15例正常人的血液作為對(duì)照組。實(shí)驗(yàn)組按WHO分級(jí),將I級(jí)和II級(jí)膠質(zhì)瘤患者的血液標(biāo)本歸類為低級(jí)別膠質(zhì)瘤組,將III級(jí)和IV級(jí)膠質(zhì)瘤患者血液標(biāo)本歸類為高級(jí)別膠質(zhì)瘤組。通過RT-qPCR法分別檢測(cè)miRNA-21、miRNA-221、miRNA-222在人腦膠質(zhì)瘤患者血液和正常人血液中的表達(dá)量;統(tǒng)計(jì)分析miRNA-21、miRNA-221及miRNA-222在實(shí)驗(yàn)組與對(duì)照組、高級(jí)別膠質(zhì)瘤和低級(jí)別膠質(zhì)瘤組之間的表達(dá)差異。 結(jié)果: 1.miRNA-21、miRNA-221、miRNA-222在膠質(zhì)瘤患者血液中的表達(dá)水平比正常人血液中的表達(dá)均有升高。 2.miRNA-21在高級(jí)別膠質(zhì)瘤患者和低級(jí)別膠質(zhì)瘤患者血液中的表達(dá)量對(duì)比正常人血液中的miRNA-21表達(dá)量均有明顯統(tǒng)計(jì)學(xué)差異(P0.05而且三組之間兩兩比較表達(dá)差異均有統(tǒng)計(jì)學(xué)學(xué)意義)。 3.miRNA-221在高級(jí)別膠質(zhì)瘤血清的表達(dá)量與正常人群和低級(jí)別膠質(zhì)瘤患者相比均有統(tǒng)計(jì)學(xué)差異(P0.05),但在低級(jí)別膠質(zhì)瘤與正常人群相比無統(tǒng)計(jì)學(xué)差異(P0.05)。 4. miRNA-222僅在高級(jí)別膠質(zhì)瘤組患者血清中表達(dá)量與正常人組中的對(duì)比表達(dá)有統(tǒng)計(jì)學(xué)差異(P0.05),低級(jí)別膠質(zhì)瘤組與正常人群以及高級(jí)別膠質(zhì)瘤組與低級(jí)別膠質(zhì)瘤組差異較小,無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論: 1.miRNA-21、miRNA-221、miRNA-222在膠質(zhì)瘤患者的血清中均有較高的表達(dá),提示這三者均可參與膠質(zhì)瘤的形成。 2.miRNA-21在高級(jí)別膠質(zhì)瘤組和低級(jí)別膠質(zhì)瘤組患者以及正常人組血漿中有差異性表達(dá),提示其可能參與了膠質(zhì)瘤的形成、發(fā)展、惡化過程。 3.miRNA-221、miRNA-222在高級(jí)別膠質(zhì)瘤組血清中表達(dá)較高,提示其在高級(jí)別膠質(zhì)瘤形成過程中可能起著重要作用。 4.miRNA-222在高級(jí)別膠質(zhì)瘤組與低級(jí)別膠質(zhì)瘤病人組血液中的表達(dá)差異無統(tǒng)計(jì)學(xué)意義,提示miRNA-221、miRNA-222、miRNA-21表達(dá)失衡可能導(dǎo)致膠質(zhì)瘤的惡化成為高級(jí)別膠質(zhì)瘤。 5.血液中檢測(cè)miRNA-21、miRNA-221、miRNA-222為膠質(zhì)瘤的早期無創(chuàng)診斷及其分級(jí)和預(yù)后評(píng)估提供了借鑒意義。
[Abstract]:Objective: to detect the expression of miRNA-21mRNA-221mRNA-222 in the blood of glioma patients and to explore the relationship between miRNA-21mRNA-221 miRNA-222 and the occurrence and development of glioma, which provides a feasible method and approach for the early diagnosis and progression of glioma.Methods: the blood samples of 60 patients with glioma confirmed by pathology were collected as experimental group and 15 normal persons as control group.According to WHO grade, the blood samples of grade I and grade II gliomas were classified as low-grade gliomas, and those of grade III and grade IV gliomas were classified as high-grade gliomas.The expression of miRNA-21mRNA-221mRNA-222 in the blood of human glioma patients and normal people was detected by RT-qPCR method, and the difference of miRNA-21 miRNA-221 and miRNA-222 expression between the experimental group and control group, high grade glioma group and low grade glioma group was analyzed statistically.Results:1. The expression of miRNA-21mRNA-221 miRNA-222 in the blood of glioma patients was higher than that in normal controls.The expression of 2.miRNA-21 in the blood of high grade glioma patients and low grade glioma patients was significantly different compared with that of normal controls (P0.05) and the differences between the three groups were statistically significant.The expression of 3.miRNA-221 in the serum of high grade glioma was significantly different from that of normal and low grade glioma patients (P 0.05), but there was no significant difference between low grade glioma and normal group.4. The expression of miRNA-222 in the serum of patients with high grade glioma was significantly different from that of normal controls (P 0.05), but the difference between low grade glioma group and normal group and between high grade glioma group and low grade glioma group was smaller than that in high grade glioma group and low grade glioma group.There was no statistical significance (P 0.05).Conclusion:1. MiRNA-21mRNA-221 miRNA-222 was highly expressed in the serum of glioma patients, suggesting that all of them were involved in the formation of glioma.The differential expression of 2.miRNA-21 in the plasma of patients with high grade glioma, low grade glioma and normal controls suggests that 2.miRNA-21 may be involved in the formation, development and deterioration of glioma.3. MiRNA-221 miRNA-222 is highly expressed in the serum of high grade gliomas, suggesting that miRNA-222 may play an important role in the formation of high grade gliomas.There was no significant difference in the expression of 4.miRNA-222 between high grade glioma group and low grade glioma patient group, suggesting that the imbalance of miRNA-221 miRNA-222 miRNA-21 might lead to the deterioration of glioma into high grade glioma.5.The detection of miRNA-21mRNA-221 miRNA-222 in blood provides a reference for early noninvasive diagnosis, grading and prognostic evaluation of gliomas.
