LUC7L3與Ki-67在腦膜瘤組織中的表達(dá)及意義

【摘要】:目的:對腦膜瘤組織中LUC7L3和Ki-67蛋白的表達(dá)情況進(jìn)行檢測和對比,對兩種蛋白質(zhì)在腦膜瘤的發(fā)生和發(fā)展中可能的作用機(jī)制進(jìn)行探索,為腦膜瘤的診斷與治療提供新的思路,從蛋白表達(dá)水平探索腦膜瘤的作用蛋白表達(dá)情況。方法:選擇2007-2015年之間在南昌大學(xué)附屬醫(yī)院神經(jīng)外科進(jìn)行腦膜瘤切除手術(shù)的患者50例,另取20例正常腦膜組織作為對照,取自頭外傷后行內(nèi)外減壓術(shù)的患者。將手術(shù)切除的組織進(jìn)行病理學(xué)檢測,并使用MaxVision TM免疫組化法檢測LUC7L3蛋白以及Ki-67蛋白的表達(dá)情況。結(jié)合腦膜瘤病例的臨床資料,對免疫組織化學(xué)結(jié)果進(jìn)行分析、討論。結(jié)果:(1)LUC7L3在不同等級的腦膜瘤組織中都有較高的陽性檢出率(86%),但是在正常腦組織中LUC7L3的表達(dá)均為陰性,正常腦膜組織和腦膜瘤組織中LUC7l3的表達(dá)差異有顯著性(?2=28.486,p0.05),并且研究結(jié)果顯示LUC7L3的陽性率隨腫瘤組織的惡性程度增加而升高,I級腦膜瘤組織的LUC7L3陽性率為71.4%,II級腦膜瘤組織的LUC7L3的陽性率為94%,III級腦膜瘤組織的LUC7L3的陽性率為100%,其陽性程度之間的差異有統(tǒng)計(jì)學(xué)意義(?2=21.45,p0.05)。(2)Ki-67在正常腦膜中表達(dá)陽性率低(LI1%),在腦膜瘤組織中表達(dá)陽性率顯著升高,差異有統(tǒng)計(jì)學(xué)意義(?2=25.486,p0.05),Ki-67的與腫瘤組織的惡性程度也有正相關(guān)關(guān)系,本次研究結(jié)果顯示,I級腦膜瘤組織的Ki-67標(biāo)記指數(shù)平均為1%(0.00%~3.00%),II級腦膜瘤組織的Ki-67標(biāo)記指數(shù)平均為3%(2.00%~4.25%),III級腦膜瘤組織的Ki-67的標(biāo)記指數(shù)平均為6%(4.00%~20.00%),三組標(biāo)記指數(shù)具有顯著的統(tǒng)計(jì)學(xué)差異(?2=22.496,p0.01)(3)本次通過對數(shù)據(jù)進(jìn)行Spearman相關(guān)性分析,分析結(jié)果顯示LUC7L3與Ki-67值呈正相關(guān)關(guān)系(p0.05,r=0.845),并且結(jié)果顯示LUC7L3與Ki-67的表達(dá)情況與腦膜瘤的病例分級有較大的相關(guān)性,與性別、年齡、腫瘤大小、發(fā)病部位無相關(guān)性(p均0.05)。結(jié)論:(1)LUC7L3和Ki-67在腦膜瘤組織中表達(dá)的陽性率及與腫瘤的惡性程度都有較高的相關(guān)關(guān)系,在正常的腦組織中很少發(fā)現(xiàn)LUC7L3和Ki-67的表達(dá),說明LUC7L3和Ki-67的表達(dá)可能與腦膜瘤的發(fā)生和發(fā)展有密切聯(lián)系;(2)LUC7L3和Ki-67在腦膜瘤組織中的陽性表達(dá)率都較高,并且隨著腦膜瘤惡性程度的升高而升高,與腦膜瘤的惡性程度具有正相關(guān)關(guān)系,檢測這兩類蛋白量可以在腦膜瘤臨床病理學(xué)分級中作為重要的參考指標(biāo);(3)LUC7L3當(dāng)前研究較少,但是本次研究證明了LUC7L3與腦膜瘤發(fā)生和發(fā)展有正相關(guān)關(guān)系,所以可以通過抑制LUC7L3的表達(dá)抑制腦膜瘤的發(fā)生和發(fā)展。進(jìn)一步研究LUC7L3與Ki-67在腦膜瘤發(fā)生、發(fā)展中的作用機(jī)制,可能為腦膜瘤的治療提供新的途徑。
[Abstract]:Objective: to detect and compare the expression of LUC7L3 and Ki-67 proteins in meningiomas, and to explore the possible mechanism of the two proteins in the occurrence and development of meningiomas, so as to provide a new idea for the diagnosis and treatment of meningiomas.To explore the expression of functional protein in meningioma from the level of protein expression.Methods: from 2007 to 2015, 50 patients underwent meningioma resection in the neurosurgery department of Nanchang University affiliated Hospital, and another 20 normal meningeal tissues were taken from the patients undergoing internal and external decompression after head trauma.The expression of LUC7L3 protein and Ki-67 protein were detected by MaxVision TM immunohistochemical method.Combined with the clinical data of meningioma cases, the immunohistochemical results were analyzed and discussed.Results the positive rate of LUC7L3 was higher in meningiomas of different grades, but the expression of LUC7L3 was negative in normal brain tissues.There was significant difference in the expression of LUC7l3 between normal meningeal tissue and meningioma tissue. The results showed that the positive rate of LUC7L3 increased with the increase of malignant degree of tumor tissue. The positive rate of LUC7L3 in grade I meningioma was 71.4%.The positive rate of LUC7L3 was 940.The positive rate of LUC7L3 in the meningioma of grade III was 100. The difference of the positive degree was statistically significant. The positive rate of LUC7L3 in normal meninges was lower than that in normal meninges. The positive rate of LUC7L3 in meningiomas was significantly higher than that in meningiomas.There was a positive correlation between Ki-67 and malignant degree of tumor tissue.The results of this study showed that the average Ki-67 labeling index of grade I meningioma tissue was 1: 0. 00 and the Ki-67 labeling index of grade II meningioma tissue was 3 ~ 2. 00 on average, the Ki-67 labeling index of meningioma tissue of grade I was 6 ~ 4. 00 on average, and the labeling index of Ki-67 in the three groups was significant.This time, the Spearman correlation analysis of the data was carried out.The results showed that there was a positive correlation between LUC7L3 and Ki-67, and the expression of LUC7L3 and Ki-67 was significantly correlated with the grade of meningioma, but not with sex, age, tumor size and location of meningioma.Conclusion the positive expression rate of LUC7L3 and Ki-67 in meningioma tissues and the degree of malignancy of meningiomas are highly correlated, and the expression of LUC7L3 and Ki-67 in normal brain tissues is rarely found.It is suggested that the expression of LUC7L3 and Ki-67 may be closely related to the occurrence and development of meningioma. The positive rates of LUC7L3 and Ki-67 in meningioma tissues are higher than those in meningioma tissues, and increase with the increase of malignant degree of meningioma.There is a positive correlation between the malignant degree of meningioma and the detection of these two kinds of proteins, which can be used as an important reference index in the clinicopathological grading of meningioma.But this study proves that LUC7L3 has a positive correlation with the occurrence and development of meningioma, so it can inhibit the occurrence and development of meningioma by inhibiting the expression of LUC7L3.Further study on the role of LUC7L3 and Ki-67 in the pathogenesis and development of meningioma may provide a new approach for the treatment of meningioma.
