【摘要】:目的:研究VE-cadherin與N-cadherin在膠質(zhì)瘤中的表達(dá)及其與膠質(zhì)瘤相關(guān)特性的關(guān)系,探討兩者在膠質(zhì)瘤發(fā)生發(fā)展中的作用,為膠質(zhì)瘤分子治療提供理論依據(jù)。方法:①選擇瀘州醫(yī)學(xué)院附屬醫(yī)院神經(jīng)外科2011年1月—2013年1月部分膠質(zhì)瘤手術(shù)切除后病理科存放的優(yōu)質(zhì)膠質(zhì)瘤病理標(biāo)本65例,所有的患者術(shù)前身體健康,選擇18例顱腦外傷后行內(nèi)外減壓術(shù)患者的正常腦組織作為對(duì)照。②所獲得膠質(zhì)瘤組織及正常腦組織先行HE染色,再加用免疫組織化學(xué)技術(shù),按照世界衛(wèi)生組織(WHO)2007年中樞神經(jīng)系統(tǒng)腫瘤分型、分級(jí)標(biāo)準(zhǔn)對(duì)膠質(zhì)瘤進(jìn)行分類。③標(biāo)本行免疫組織化學(xué)染色后檢測(cè)正常腦組織及腫瘤組織中VE-cadherin、N-cadherin的表達(dá)情況。④分析兩指標(biāo)在正常腦組織及膠質(zhì)瘤中的表達(dá)情況,將指標(biāo)表達(dá)強(qiáng)度進(jìn)行分類。⑤從四個(gè)方面分析兩指標(biāo)的表達(dá)程度:正常腦組織和膠質(zhì)瘤組織中的表達(dá)情況;在正常腦組織及不同級(jí)別膠質(zhì)瘤組織中的表達(dá)情況;不同病理類型中膠質(zhì)瘤組織的表達(dá)情況;在合并有不同程度瘤周水腫的膠質(zhì)瘤組織中表達(dá)情況。然后進(jìn)行統(tǒng)計(jì)分析。結(jié)果:①VE-cadherin在膠質(zhì)瘤組織和正常腦組織中表達(dá)之間比較差異有顯著性(Z=-3.640;P=0.0000.005),VE-cadherin在膠質(zhì)瘤組織中表達(dá)高于正常腦組織。②VE-cadherin在正常腦組織、Ⅰ級(jí)、Ⅱ級(jí)、Ⅲ級(jí)和Ⅳ膠質(zhì)瘤級(jí)之間表達(dá)比較差異有顯著性(x2=22.181, P=0.0000.05), VE-cadherin表達(dá)強(qiáng)度與膠質(zhì)瘤級(jí)別呈正相關(guān)(rs=0.491,P=0.0000.05)。③不同病理類型的膠質(zhì)瘤組織中,VE-cadherin表達(dá)差異無(wú)顯著性(x2=11.289, P=0.1260.005)。④VE-cadherin在合并無(wú)水腫、輕度水腫、中度水腫和重度水腫膠質(zhì)瘤中表達(dá)差異有顯著性(x2=21.257, P=0.0000.008), VE-cadherin表達(dá)強(qiáng)度與膠質(zhì)瘤瘤周水腫程度呈正相關(guān)(r,=0.504, P=0.0000.05)。(?) N-cadherin在正常腦組織與膠質(zhì)瘤組織中的表達(dá)程度差異有顯著性(Z=-4.320, P=0.0000.05), N-cadherin在膠質(zhì)瘤組織中的表達(dá)高于正常腦組織。⑥N-cadherin在正常腦組織、Ⅰ級(jí)、Ⅱ級(jí)、Ⅲ工級(jí)和工Ⅳ膠質(zhì)瘤級(jí)之間表達(dá)比較差異有顯著性(x2=35.246, P=0.0000.05), N-cadherin表達(dá)強(qiáng)度與膠質(zhì)瘤級(jí)別呈正相關(guān)(rs=0.614, P=0.0000.05)。⑦在不同組織病理類型的膠質(zhì)瘤中,N-cadherin表達(dá)差異無(wú)顯著性(x2=11.958,P=0.1020.05)⑧N-cadherin在合并無(wú)水腫、輕度水腫、中度水腫和重度水腫膠質(zhì)瘤中表達(dá)強(qiáng)度差異有顯著性(x2=17.440,P=0.0010.05),N-cadherin表達(dá)強(qiáng)度與膠質(zhì)瘤瘤周水腫程度呈正相關(guān)(rs=0.454,P=0.0000.05)。⑨VE-cadherin與N-cadherin在膠質(zhì)瘤組織中的表達(dá)呈正相關(guān)(rs=0.206,P=0180.05)。結(jié)論:②VE-cadherin、 N-cadherin蛋白在膠質(zhì)瘤組織和正常腦組織中表達(dá)差異有顯著性,兩者在膠質(zhì)瘤組織中的表達(dá)均高于正常腦組織,且隨著膠質(zhì)瘤分級(jí)的增高而增高。提示他們協(xié)同通過(guò)某機(jī)制參與膠質(zhì)瘤發(fā)生發(fā)展,影響膠質(zhì)瘤的惡性程度。②VE-cadherin、 N-cadherin在不同膠質(zhì)瘤組織病理類型中的表達(dá)差異無(wú)顯著性,提示兩者與膠質(zhì)瘤的組織病理類型無(wú)關(guān),也可能是本實(shí)驗(yàn)樣本量小或者是選擇偏倚所致,需進(jìn)一步研究。③對(duì)VE-cadherin、N-cadherin在合并不同程度瘤周水腫膠質(zhì)瘤組織中的表達(dá)研究發(fā)現(xiàn),瘤周水腫越嚴(yán)重,兩者的表達(dá)越明顯,提示VE-cadherin、N-cadherin參與膠質(zhì)瘤瘤周水腫的發(fā)生發(fā)展,加重膠質(zhì)瘤病情。④對(duì)VE-cadherin與N-cadherin在膠質(zhì)瘤組織中的表達(dá)相關(guān)性研究發(fā)現(xiàn),VE-cadherin與N-cadherin在膠質(zhì)瘤組織中的表達(dá)呈正相關(guān),兩者可能在膠質(zhì)瘤組織中協(xié)同促進(jìn)膠質(zhì)瘤血管生成,影響膠質(zhì)瘤的發(fā)生發(fā)展。
[Abstract]:Objective: To investigate the relationship between the expression of VE-cadherin and N-cadherin in gliomas and its correlation with glioma characteristics, discuss their role in the development of glioma, and provide a theoretical basis for the treatment of glioma. Methods: select the Department of Neurosurgery of Affiliated Hospital of Luzhou Medical College from January 2011 to January 2013 was part of glioma after surgery the pathology department store quality glioma 65 cases were all patients with preoperative health, 18 cases of craniocerebral trauma underwent decompressive craniotomy patients of normal brain tissue as control. The acquired glioma tissue and normal brain tissue to HE staining, and immunohistochemistry, in accordance with the WHO (WHO) 2007 central nervous system tumor type, the glioma classification standard. The specimens with immunohistochemical staining after detection of normal brain tissue and tumor tissue VE-cadherin, N-cadherin expression. Then analyze the results. Results: VE-cadherin had significant difference between the expression in glioma tissues and normal brain tissues (Z=-3.640; P=0.0000.005). The expression of VE-cadherin in glioma tissues than in normal tissues. The VE-cadherin in normal brain tissue, grade I, grade II, there are significant differences between the expression of glioma grade III and IV A (x2=22.181, P=0.0000.05), the expression of VE-cadherin was positively correlated with the level of glioma (rs=0.491, P=0.0000.05).
