【摘要】：目的：探討垂體腺瘤中MUC1的表達(dá)與垂體腺瘤侵襲性的關(guān)系。 方法:采用免疫組化方法檢測26例侵襲性垂體腺瘤和29例非侵襲性垂體腺瘤組織中MUC1的表達(dá)。將55例垂體腺瘤組織的標(biāo)本全部經(jīng)10%甲醛液固定后，進(jìn)行石蠟包埋后，，再以5μm連續(xù)切片。做好切片常規(guī)給予脫蠟、水化后，用內(nèi)源性過氧化物酶阻斷劑和小牛血清分別在室溫下孵育30分鐘，以減低非特異性染色，排除干擾。檢測標(biāo)本均經(jīng)微波法抗原修復(fù)。MUC1抗體（1:100）4攝氏度孵育過夜；生物素標(biāo)記的二抗和抗生物素過氧化物酶連接物室溫分別孵育30分鐘；DAB顯色1-3分鐘；蘇木素襯染，脫水、透明、封片、光鏡觀察。 結(jié)果:侵襲性垂體腺瘤(IPA)組織中可見MUC1強(qiáng)陽性表達(dá)，陽性表達(dá)為深棕色、棕黃色顆粒，陽性信號主要位于細(xì)胞膜及胞漿內(nèi)；非侵襲性垂體腺瘤(NIPA)可見弱陽性反應(yīng)信號或未見MUC1表達(dá)。MUC1在侵襲性垂體腺瘤組織中的陽性表達(dá)率與非侵襲性垂體腺瘤組織之間存在顯著性差異（分別76.9％和27.6%，P0.01）。 結(jié)論: MUC1在侵襲性垂體腺瘤（IPA）組織中的陽性表達(dá)率與非侵襲性垂體腺瘤(NIPA)組織之間存在顯著性差異，說明了MUC1在垂體腺瘤（PA）形成中可能有強(qiáng)烈的致瘤作用，并和腫瘤的侵襲性生長密切相關(guān)。因此檢測MUC1在PA組織中的表達(dá)，有可能成為判斷IPA的一種新的腫瘤標(biāo)志物，對于臨床醫(yī)師診判斷垂體腺瘤侵襲性、治療以及預(yù)后判斷等都有會非常大的幫助。
[Abstract]:Objective : To investigate the relationship between expression of MUC1 in pituitary adenoma and invasion of pituitary adenoma .

Methods : Immunohistochemical method was used to detect the expression of MUC1 in 26 patients with invasive pituitary adenoma and 29 non - invasive pituitary adenoma .
the biotin - labeled di - and anti - biotin peroxidase linker were incubated at room temperature for 30 minutes , respectively ;
DAB is developed for 1 - 3 minutes ;
Subalpine lining dyeing , dehydration , transparency , sealing , light microscope observation .

Results : The positive expression of MUC1 was found in invasive pituitary adenoma ( IPA ) tissue , and the positive expression was dark brown and brown - yellow particles . The positive signal was mainly located in the cell membrane and cytoplasm .
Non - invasive pituitary adenoma ( NIPA ) showed weak positive response signal or no MUC1 expression . There was a significant difference between positive expression rate and non - invasive pituitary adenoma tissue in invasive pituitary adenoma ( 76.9 % vs 27 . 6 % , P0.01 ) .

Conclusion : The positive expression rate of MUC1 in invasive pituitary adenoma ( IPA ) tissue is significantly different from non - invasive pituitary adenoma ( NIPA ) .
