音樂(lè)對(duì)大鼠顳葉癲癇治療作用的初步探究

【摘要】:癲癇是一種常見的中樞神經(jīng)系統(tǒng)慢性疾病。雖然臨床上已有多種治療手段,但它們的有效性和實(shí)用性受到副作用、嚴(yán)重并發(fā)癥、手術(shù)風(fēng)險(xiǎn)以及昂貴的費(fèi)用等因素的限制。因此尋求更加安全、有效、并且經(jīng)濟(jì)上可承受的癲癇治療/輔助治療手段成為一項(xiàng)亟待的任務(wù)。本實(shí)驗(yàn)采用氯化鋰-匹羅卡品癲癇大鼠模型來(lái)探究音樂(lè)對(duì)癲癇的治療作用和及其神經(jīng)機(jī)制。實(shí)驗(yàn)采用72只Wistar大鼠,隨機(jī)分成4組:音樂(lè)組(n=21)、白噪音組(n=21)、癲癇對(duì)照組(n=21)和健康對(duì)照組(nn=9)。氯化鋰-匹羅卡品注射過(guò)的大鼠一旦呈現(xiàn)癲癇持續(xù)發(fā)作狀態(tài)后,即開始對(duì)音樂(lè)組和白噪音組分別播放莫扎特K488音樂(lè)和白噪音(75分貝,每天21點(diǎn)至次日9點(diǎn)連續(xù)播放12小時(shí)),共持續(xù)9周。采用24小時(shí)連續(xù)行為視頻觀察和EEG記錄等手段,觀察音樂(lè)環(huán)境對(duì)癲癇的影響。實(shí)驗(yàn)結(jié)果表明,音樂(lè)治療6周后,音樂(lè)組的癲癇發(fā)作頻率比癲癇組低78.33±7.01%(n=7),發(fā)作持續(xù)時(shí)間短33.65±4.45%(n=7),癲癇發(fā)作等級(jí)降低22.28±9.93%(n=7),發(fā)作間期棘波頻率和幅值分別降低了25.18±6.81%(n=12)和12.58±3.71%(n=12)。此外,音樂(lè)對(duì)癲癇動(dòng)物所表現(xiàn)出的認(rèn)知功能障礙也有顯著的改善作用。在新異物體識(shí)別實(shí)驗(yàn)中,癲癇組大鼠對(duì)于新異物體的偏好顯著地(p0.05)低于健康組大鼠,其探索新異物體的時(shí)間比健康組短33.11±2.50%(n=6):而音樂(lè)組動(dòng)物的新異物體探索時(shí)間比癲癇組增長(zhǎng)了28.57±7.18%(nn=7),達(dá)到了與健康組相同的水平(p0.05)。此外,音樂(lè)還可能具有緩解癲癇動(dòng)物焦慮行為的作用。曠場(chǎng)實(shí)驗(yàn)中,癲癇組在中央?yún)^(qū)域活動(dòng)時(shí)間顯著高于健康組,而音樂(lè)組的這一指標(biāo)呈現(xiàn)降低的趨勢(shì),雖然與癲癇組相比統(tǒng)計(jì)學(xué)上未達(dá)到顯著性水平(p=0.059)。我們還發(fā)現(xiàn),音樂(lè)可以提高抗癲癇藥物拉莫三嗪的藥效。我們的實(shí)驗(yàn)結(jié)果為音樂(lè)對(duì)癲癇及其所導(dǎo)致的認(rèn)知功能障礙的治療效果提供了充分的實(shí)驗(yàn)證據(jù),對(duì)于治療成人癲癇具有十分重要的臨床意義。
[Abstract]:Epilepsy is a common chronic disease of the central nervous system. Although there are many kinds of clinical treatment, but effective and practical by their side effects, complications, operative risk and high cost and other factors. Therefore, seeking more secure, effective, and affordable treatment of epilepsy / adjuvant therapy means to become a key task. This experiment using lithium chloride pilocarpine induced epilepsy rat model to explore the effects of music therapy on epilepsy and its neural mechanism. The 72 Wistar rats were randomly divided into 4 groups: the music group (n=21), white noise group (n=21), epilepsy control group (n=21) and control group (nn=9). Lithium pilocarpine injected rats once showed seizures after the start of the music group and white noise group were playing Mozart K488 music and white noise (75 dB per. Day 21 to 9 the next day, 12 hours of continuous playback) lasted 9 weeks. The 24 hours of continuous observation and EEG behavior video recording and other means to observe the effect of environmental music on epilepsy. The experimental results show that the music after 6 weeks of treatment, the music group the seizure frequency of epilepsy group than low (n=7 78.33 + 7.01%), short duration of 33.65 + 4.45% attack (n=7), seizure reduced the levels of 22.28 + 9.93%, 25.18 + 6.81% (n=7) to reduce the interictal spike wave frequency and amplitude respectively (n=12) and 12.58 + 3.71% (n=12). In addition, the music show on the epilepsy animal cognitive dysfunction also improved significant effect. In the novel object recognition experiment, epilepsy rats for novel object preference (P0.05) was significantly lower than that of healthy rats, the time to explore the novel object than the healthy group 33.11 + 2.50% (n=6): new and different objects to explore the music group animal than epilepsy group Growth of 28.57 + 7.18% (nn=7), at the same level with the healthy group (P0.05). In addition, the music may also can alleviate the epilepsy animal anxiety behavior role. The open field test, the epilepsy group in the central area of activity time was significantly higher than that in healthy group, and the music group the index showed a decreasing trend although, compared with the epilepsy group did not reach statistical significant level (p=0.059). We also found that music can improve the efficacy of antiepileptic drug lamotrigine. Our experimental results provide experimental evidence for music on cognitive dysfunction caused by epilepsy and its treatment effect, it has very important clinical significance for treatment adult epilepsy.
