大鼠腦缺血再灌注后海馬組織中GFAP和p-STAT3表達(dá)研究

【摘要】:目的檢測(cè)腦缺血再灌注(I/R)后海馬齒狀回區(qū)(dentate gyrus,DG)星形膠質(zhì)細(xì)胞活化增殖及其磷酸化信號(hào)轉(zhuǎn)導(dǎo)與轉(zhuǎn)錄激活子3(phosphorylated STAT3,p-STAT3)表達(dá)情況,為調(diào)控星形膠質(zhì)細(xì)胞異;罨鲋程峁├碚撘罁(jù)。方法線栓法制作大鼠腦局部I/R模型,48只Sprague-Dawley(SD)大鼠隨機(jī)分為假手術(shù)(Sham)組、I/R-24 h組、I/R-72 h組及I/R-7 d組。免疫熒光法及Western印跡檢測(cè)缺血后不同時(shí)間點(diǎn)患側(cè)海馬區(qū)膠質(zhì)原纖維酸性蛋白(glial fibrillary acidic protein,GFAP)標(biāo)記陽(yáng)性細(xì)胞及p-STAT3表達(dá)情況。結(jié)果與Sham組比較,I/R-72 h和I/R-7 d組海馬區(qū)GFAP蛋白表達(dá)增加(P0.05);隨缺血時(shí)間延長(zhǎng),GFAP蛋白表達(dá)增加呈上升趨勢(shì),且24 h、72 h和7 d各時(shí)間點(diǎn)之間兩兩比較差異均具有統(tǒng)計(jì)學(xué)意義(P0.05)。與Sham組比較,腦I/R后海馬區(qū)各時(shí)間點(diǎn)p-STAT3表達(dá)量均顯著增加(P0.05);p-STAT3表達(dá)在24 h最高,72 h開(kāi)始下降,7 d仍有一定量表達(dá),與I/R-24 h組比較,I/R-72 h組和I/R-7 d組p-STAT3表達(dá)量均顯著降低(P0.05)。結(jié)論腦I/R損傷后,海馬區(qū)星形膠質(zhì)細(xì)胞大量增殖及其p-STAT3表達(dá)增加;且p-STAT3的表達(dá)高峰在時(shí)間上早于星形膠質(zhì)細(xì)胞的大量增殖。
[Abstract]:Objective to detect the activation and proliferation of astrocytes and the expression of phosphorylated signal transduction and activator 3(phosphorylated STAT3p-STAT3 in dentate gyrus of dentate gyrus after cerebral ischemia-reperfusion (I / R) so as to provide a theoretical basis for regulating the abnormal activation and proliferation of astrocytes.Methods 48 Sprague-Dawley SD rats were randomly divided into I / R 72 h group and I/R-7 d group.The expression of glial fibrillary acidic protein (p-STAT3) and glial fibrillary acidic protein (glial fibrillary acidic protein) positive cells were detected by immunofluorescence and Western blotting at different time points after ischemia.Results compared with Sham group, the expression of GFAP protein increased in hippocampus of I- / R-72 h group and I/R-7 d group, and the expression of GFAP protein increased with the prolongation of ischemia time, and there were significant differences between the two groups at 24 h and 7 d.涓嶴ham緇勬瘮杈,
