病灶區(qū)灌注羅格列酮對腦出血家兔血腫周圍咬合蛋白和閉合小環(huán)蛋白1 mRNA表達水平及血-腦屏障通透性的影響

【摘要】:目的觀察病灶區(qū)灌注羅格列酮(RSG)對家兔腦出血(ICH)模型血腫周圍咬合蛋白和閉合小環(huán)蛋白1(ZO-1)mRNA表達水平、血-腦屏障(BBB)通透性及神經(jīng)功能評分的影響。方法選擇健康雄性家兔45只,體質(zhì)量2.0~2.5 kg,按隨機數(shù)字表法分為對照組、ICH模型組和羅格列酮(RSG)治療組,每組15只(其中5只用于BBB測定)。對照組模擬制作顱內(nèi)血腫的過程,穿刺成功后對兔靶點注射等滲鹽水0.3 ml,6 h后再次注入等滲鹽水0.1 ml;ICH模型組穿刺成功后靶點注射自體非抗凝動脈血0.3 ml,6 h后靶點再次注入等滲鹽水0.1 ml;RSG治療組在ICH模型制作成功后6 h,血腫區(qū)灌注RSG 0.5 mg(溶于0.1 ml等滲鹽水中)。繼續(xù)飼養(yǎng)至第7天,對各組家兔進行神經(jīng)功能缺損評分(Purdy評分)后處死,逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(RT-PCR)法檢測血腫周圍腦組織咬合蛋白及ZO-1 mRNA的表達水平;采用甲酰胺方法測量血腫周圍腦組織伊文思藍(EB)含量以評估BBB通透性。結(jié)果 (1)神經(jīng)功能評分:對照組、ICH模型組、RSG治療組Purdy評分分別為(2.53±0.05)、(8.13±0.06)、(6.67±0.08)分,組間差異有統(tǒng)計學(xué)意義(F=459.116,P0.01);與對照組比較,ICH模型組、RSG治療組的Purdy評分均明顯增加(均P0.01);與ICH模型組比較,RSG治療組的Purdy評分降低(P0.05);(2)咬合蛋白及ZO-1 mRNA表達水平:對照組、ICH模型組、RSG治療組咬合蛋白及ZO-1 mRNA差異均有統(tǒng)計學(xué)意義(每組分別為1.013±0.051、1.001±0.045;0.221±0.017、0.247±0.019;0.498±0.041、0.613±0.045;F值分別為443.924、381.929,均P0.01),與對照組比較,ICH模型組、RSG治療組咬合蛋白及ZO-1 mRNA表達均顯著減少(均P0.01);與ICH模型組比較,RSG治療組咬合蛋白及ZO-1 mRNA的表達增加(均P0.05);(3)BBB通透性:對照組、ICH模型組、RSG治療組EB含量分別為(12.0±1.0)、(51.6±0.9)、(36.4±1.0)μg/g,組間差異有統(tǒng)計學(xué)意義(F=223.516,P0.01);與對照組比較,ICH模型組、RSG治療組的EB含量明顯增加(均P0.01);與ICH模型組比較,RSG治療組EB含量明顯降低(P0.01)。結(jié)論病灶區(qū)灌注RSG可明顯改善家兔ICH后的神經(jīng)功能,增加血腫周圍腦組織咬合蛋白和ZO-1mRNA的表達水平,降低BBB通透性。
[Abstract]:Objective to observe the effects of rosiglitazone RSGs on the expression of occlusal protein and closed microannular protein 1a ZO-1mRNA, BBB permeability and neurological function score of intracerebral hemorrhage (ICH) model in rabbits. Methods 45 healthy male rabbits were selected. The rats were randomly divided into two groups: control group (n = 15) and rosiglitazone group (n = 15). The control group was used to simulate the process of making intracranial hematoma. After successful puncture, rabbit target was injected with isoosmotic saline 0.3 ml / h and then re-injected 0.1 ml / ml ICH after puncture. In the model group, the target was injected with autogenous non-anticoagulant arterial blood 0.3 ml / min 6 h later, then the target was re-injected with 0.1 ml / L isoosmotic saline in ICH group. 6 hours after the model was successfully made, RSG 0.5 mg (dissolved in 0.1 ml isoosmotic saline) was infused into the hematoma area. The rabbits in each group were killed after the neurological impairment score (Purdy score). The expression of occlusal protein and ZO-1 mRNA were detected by reverse transcriptase polymerase chain reaction (RT-PCR) in the brain tissue around the hematoma. The BBB permeability was evaluated by formamide method. Results: the Purdy score of the control group was 2.53 鹵0.05 鹵0.05, and the Purdy score was 6.67 鹵0.08 in the control group. There was significant difference between the two groups (P < 0.01); compared with the control group, the Purdy score of the treated group increased significantly (all P 0.01; compared with the ICH model group, the Purdy score of the Purdy group decreased the level of P0.05P0.05) and the expression of ZO-1 mRNA in the control group was significantly higher than that in the control group. The occlusal protein and ZO-1 mRNA in the ICH model group were significantly lower than those in the control group (1.013 鹵0.051 鹵1.001 鹵0.045 鹵0.221 鹵0.017) 0.247 鹵0.041 鹵0.498 鹵0.041 鹵0.613 鹵0.045 F, respectively (P 0.01). Compared with the control group, the occlusal protein and ZO-1 mRNA expression in the ICH model group were significantly decreased (P 0.01). The expression of occlusal protein and ZO-1 mRNA in the ICH model group were significantly higher than those in the control group (P0.05). The EB content in the control group was 12.0 鹵1.0 鹵1.0 鹵0.96 鹵0.96 鹵0.96 鹵1.0 渭 g / g, respectively. The difference between the two groups was statistically significant (P < 0.05), and that of the control group was higher than that of the control group (P < 0.05). Compared with the ICH model group, the EB content in the treatment group was significantly lower than that in the control group. Conclusion the perfusion of RSG in the focus area can obviously improve the nerve function after ICH in rabbits. The expression of occlusal protein and ZO-1mRNA were increased and the permeability of BBB was decreased.
