“養(yǎng)腦方”對永久性局灶性腦缺血大鼠恢復早期神經(jīng)功能的影響及其機制的研究

【摘要】:目的:觀察“養(yǎng)腦方”對永久性局灶性腦缺血大鼠恢復早期行為學的改變,探討其促進神經(jīng)功能恢復的作用機制;為腦梗死患者早期的臨床治療提供一種新的中藥干預方法。 方法:采用線栓法制作永久性大腦中動脈栓塞(pMCAO)模型。術后24小時采用Longa評分法評估大鼠肢體損傷程度。本研究將實驗動物完全隨機分為:假手術組、pMCAO模型組、西藥組和中藥組。以“養(yǎng)腦方”作為干預手段,術后3天至14天定時灌胃給藥,1次/天。分別于腦缺血3天、7天、10天和14天再次行Longa評分及橫木行走試驗評分。14天末次給藥后處死所有大鼠。采用HE染色觀察腦組織形態(tài)結(jié)構;運用逆轉(zhuǎn)錄-聚合酶鏈反應(RT-PCR)法檢測皮質(zhì)缺血區(qū)低氧誘導因子-1α(HIF-1α)和血管內(nèi)皮生長因子(VEGF)的轉(zhuǎn)錄水平;運用免疫組織化學染色法觀察皮質(zhì)缺血區(qū)HIF-1α、 VEGF的表達。采用完全隨機設計的多樣本方差分析和SNK-q檢驗對實驗數(shù)據(jù)進行統(tǒng)計學比較。 結(jié)果:1、與pMCAO模型組相比,“養(yǎng)腦方”對恢復早期腦缺血大鼠左側(cè)姿勢反射有顯著的改善作用;能明顯提高大鼠運動協(xié)調(diào)整合的能力(P0.01);2、中藥組和西藥組皮質(zhì)缺血區(qū)腦組織中HIF-1α、 VEGF的表達明顯高于假手術組和模型組(P=0.000,P0.01);與西藥組相比,中藥組皮質(zhì)缺血區(qū)腦組織VEGF的表達高(P=0.009,P0.01);3、皮質(zhì)缺血區(qū)腦組織HIF-1α、VEGF的轉(zhuǎn)錄水平在中藥組與西藥組中無統(tǒng)計學差異(p0.05)。 結(jié)論:“養(yǎng)腦方”能改善pMCAO腦缺血大鼠恢復早期的神經(jīng)功能,其機制可能與HIF-1α上調(diào)VEGF的表達有關。
[Abstract]:Objective: to observe the effect of "Nourishing Nao Fang" on the early behavioral changes in rats with permanent focal cerebral ischemia, to explore the mechanism of promoting the recovery of nerve function, and to provide a new Chinese medicine intervention method for the early clinical treatment of patients with cerebral infarction. Methods: the model of permanent middle cerebral artery embolization (pMCAO) was established by thread embolization. The limb injury was evaluated by Longa score 24 hours after operation. The experimental animals were randomly divided into sham operation group and pMCAO model group. Western medicine group and traditional Chinese medicine group. "Yangnao prescription" as intervention means, From 3 to 14 days after operation, the rats were given intragastric administration once a day. The Longa score and the trabecular walking test score were again performed on the 3rd day, 7th day, 10th day, and 14th day, respectively. All the rats were killed after the last administration of the drug on the 14th day. The morphology of brain tissue was observed by HE staining. The transcription levels of hypoxia inducible factor-1 偽 (HIF-1 偽) and vascular endothelial growth factor (VEGF) were detected by reverse transcription-polymerase chain reaction (RT-PCR). The expression of HIF-1 偽 and VEGF in ischemic cortex was observed by immunohistochemical staining, and the experimental data were compared statistically by means of multisample ANOVA and SNK-q test. Results compared with pMCAO model group, "Yangnao Fang" could significantly improve the left postural reflex in early cerebral ischemia rats. The expression of HIF-1 偽 and VEGF in cerebral tissue of cortex ischemia area in Chinese medicine group and western medicine group was significantly higher than that in sham operation group and model group, compared with western medicine group, the expression of HIF-1 偽 and VEGF in cerebral tissue of cerebral ischemia area in Chinese medicine group and western medicine group was significantly higher than that in sham operation group and model group. The expression of VEGF in cerebral tissue of cerebral ischemia area in Chinese medicine group was higher than that in control group. There was no significant difference in the transcription level of HIF-1 偽 -VEGF between traditional Chinese medicine group and western medicine group. Conclusion: "Nourishing Nao Fang" can improve the early recovery of neural function in rats with pMCAO cerebral ischemia, and its mechanism may be related to the up-regulation of VEGF expression by HIF-1 偽.
