神經(jīng)干細(xì)胞移植帕金森病模型大鼠行為學(xué)及對炎癥因子的調(diào)節(jié)效應(yīng)

【摘要】:背景:研究證實,干細(xì)胞移植可抑制帕金森病模型動物多巴胺能神經(jīng)元的喪失,改善其行為學(xué)癥狀,具有一定的治療作用。目的:觀察神經(jīng)干細(xì)胞移植后帕金森病大鼠行為學(xué)及炎癥因子的變化。方法:將108只SD大鼠隨機分為正常對照組、模型組及干細(xì)胞移植組,每組36只。模型組及干細(xì)胞移植組在腦內(nèi)紋狀體內(nèi)注射6-羥基多巴胺建立帕金森病模型,造模成功后即刻,干細(xì)胞移植組于紋狀體內(nèi)注射1×109 L-1的神經(jīng)干細(xì)胞懸液5μL,模型組注射等量的生理鹽水。細(xì)胞移植后1,2,4周,3組每個時間點各取大鼠6只,腹腔注射0.5 mg/kg的阿撲嗎啡,注射后10 min開始計數(shù)旋轉(zhuǎn)圈數(shù),計時5 min;ELISA法檢測腦組織內(nèi)腫瘤壞死因子α、白細(xì)胞介素1β、干擾素γ及白細(xì)胞介素4水平。結(jié)果與結(jié)論:(1)正常對照組未發(fā)生旋轉(zhuǎn)行為。干細(xì)胞移植組與模型組出現(xiàn)旋轉(zhuǎn)行為,干細(xì)胞移植組于移植后2周開始旋轉(zhuǎn)次數(shù)明顯減少,干細(xì)胞移植組移植后2,4周的旋轉(zhuǎn)次數(shù)明顯低于模型組(P0.05),且組內(nèi)移植后2,4周的旋轉(zhuǎn)次數(shù)明顯低于移植后1周(P0.05);(2)與正常對照組比較,模型組移植后不同時間點的腫瘤壞死因子α、白細(xì)胞介素1β水平明顯升高(P0.05);與模型組比較,干細(xì)胞移植組移植后不同時間點的腫瘤壞死因子α、白細(xì)胞介素1β水平明顯降低(P0.05);(3)與正常對照組比較,模型組移植后不同時間點的干擾素γ、白細(xì)胞介素4水平均明顯升高(P0.05);與模型組比較,干細(xì)胞移植組移植后1,4周的干擾素γ、白細(xì)胞介素4水平均明顯升高(P0.05);(4)結(jié)果表明,神經(jīng)干細(xì)胞移植可改善帕金森病大鼠的行為學(xué)癥狀,降低其腦內(nèi)促炎性因子腫瘤壞死因子α、白細(xì)胞介素1β水平,提高抗炎因子白細(xì)胞介素4的水平。
[Abstract]:Background: stem cell transplantation has been shown to inhibit the loss of dopaminergic neurons and improve behavioral symptoms in Parkinson's disease model animals. Objective: to observe the changes of behavioral and inflammatory factors in rats with Parkinson's disease after transplantation of neural stem cells. Methods: 108 SD rats were randomly divided into normal control group, model group and stem cell transplantation group. The model group and stem cell transplantation group were injected with 6-hydroxydopamine into the striatum to establish Parkinson's disease model. The stem cell transplantation group was injected with 1 脳 109 L-1 neural stem cell suspension (5 渭 L) in striatum and the model group was injected with the same amount of normal saline. Six rats were taken from each time point in each time point of 1: 2 and 4 weeks after cell transplantation, and the rats were injected intraperitoneally with apomorphine for 0.5 mg/kg. 10 min after injection, the number of rotating circles was counted, The levels of tumor necrosis factor 偽, interleukin 1 尾, interferon 緯 and interleukin 4 in brain tissue were measured by 5 mins Elisa. Results and conclusion: there was no rotational behavior in normal control group, but rotation behavior was observed in stem cell transplantation group and model group. The number of rotation in stem cell transplantation group decreased significantly 2 weeks after transplantation. The rotation times of the stem cell transplantation group at 2 ~ 4 weeks after transplantation were significantly lower than those of the model group (P 0.05), and the rotation times of the group 2 ~ 4 weeks after transplantation were significantly lower than those of the normal control group 1 week after transplantation. The levels of tumor necrosis factor 偽 and interleukin 1 尾 in the model group were significantly higher than those in the model group at different time points (P 0.05). The levels of tumor necrosis factor 偽 and interleukin 1 尾 in stem cell transplantation group were significantly lower than those in normal control group at different time points after transplantation. The levels of interferon 緯 and interleukin 4 at different time points in model group were significantly higher than those in model group, and the levels of interferon 緯 and interleukin 4 in stem cell transplantation group were significantly higher than those in model group at 1 and 4 weeks after transplantation. Neural stem cell transplantation can improve the behavioral symptoms of Parkinson's disease rats, reduce the levels of tumor necrosis factor 偽 and interleukin-1 尾 in the brain and increase the level of anti-inflammatory factor interleukin 4 in the brain of rats with Parkinson's disease.
