【摘要】：目的： 本研究通過免疫組化法檢測Caspase-3、HSP60蛋白在不同級別星形細胞瘤中的表達，通過觀察Caspase-3、HSP60分布與星形細胞瘤臨床病理的關系，以及Caspase-3、HSP60蛋白二者之間的相關性，探討Caspase-3和HSP60蛋白在判斷星形細胞瘤惡性程度上的作用，為腫瘤臨床診斷和治療提供分子靶點。 方法： 收集來自浙江省湖州市中心醫(yī)院病理科從2005年6月到2012年12月間診斷為星形細胞瘤的手術存檔標本共58例。按2007年WHO制定的神經(jīng)系統(tǒng)腫瘤分類和分級標準分為：WHOⅠ級4例，Ⅱ級18例，Ⅲ級19例，Ⅳ級17例。其中男性37名，女性21名，年齡7～65歲，平均34.6歲。實驗用免疫組化SP檢測法。一抗為鼠抗人Caspase-3單克隆抗體和鼠抗人HSP60單克隆抗體。檢測Caspase-3蛋白和HSP60在人腦星形細胞瘤組織中的表達情況,結合臨床病理資料進行統(tǒng)計分析，數(shù)據(jù)結果應用SPSS19.0統(tǒng)計軟件處理，根據(jù)數(shù)據(jù)類型選用非參數(shù)kraskal-wallis H檢驗及Spearman相關分析，，以P 0.05為差異有統(tǒng)計學意義。 結果： 1、Caspase-3和HSP60蛋白免疫組織化學染色陽性結果為棕黃色顆粒，Caspase-3陽性顆粒主要定位在腫瘤細胞漿內(nèi)，HSP60陽性顆粒主要定位在腫瘤細胞核和細胞漿內(nèi)。 2、隨著星形細胞瘤病理級別的增高，Caspase-3的表達強度有所減弱，WHO I～Ⅳ級星形細胞瘤的Caspase-3陽性率分別為100%、77.7%、36.8%、23.5%（P 0.01）。 3、隨著星形細胞瘤病理級別的增高，HSP60的表達強度逐漸增加，WHO I～Ⅳ級星形細胞瘤的HSP60陽性率分別為25%、72.2%、59.4%、100%（P 0.01）。 4、星形細胞瘤中，Caspase-3和HSP60的蛋白表達間存在負相關（P 0.01）。 結論： Caspase-3蛋白與HSP60蛋白的表達結果提示，兩者與星形細胞瘤的生物學行為和分化程度有關，對不同級別星形細胞瘤中Caspase-3蛋白與HSP60的表達進行聯(lián)合檢測，不但有助于判斷星形細胞瘤惡性程度，而且還有助于為腫瘤的發(fā)病機制提供理論依據(jù)。
[Abstract]:Objective:. In this study, the expression of Caspase-3 hSP60 protein in astrocytomas of different grades was detected by immunohistochemistry. The relationship between the distribution of Caspase-3 hSP60 and the clinicopathology of astrocytomas and the correlation between Caspase-3 and HSP60 protein were observed. To explore the role of Caspase-3 and HSP60 protein in judging the malignant degree of astrocytoma and to provide molecular targets for clinical diagnosis and treatment of astrocytoma. Methods:. 58 cases of astrocytoma diagnosed as astrocytoma were collected from Department of Pathology, Huzhou Central Hospital, Zhejiang Province from June 2005 to December 2012.
