早期腦出血后細胞凋亡與p75NTR、TrkA的相關(guān)性研究

【摘要】:目的:探討p75NTR、TrkA在早期人高血壓腦出血后的表達,并初步闡明與細胞凋亡的聯(lián)系。 方法:收集腦出血后血腫周圍不同部位組織標(biāo)本;取血腫表面組為A組,半暗帶組為B組,腦皮質(zhì)區(qū)組為C組,其中,A組、B組為實驗組,C組為對照組;并在A、B、C三組中將標(biāo)本按出血時間(7h、≤7h)、出血量(60ml、60ml)分組。采用免疫組化檢測p75NTR、TrkA在早期人腦出血距離血腫不同區(qū)域中的表達; Tunel方法檢測出血后各部位的凋亡細胞率;統(tǒng)計學(xué)方法分析并闡明p75NTR、TrkA及凋亡細胞在時空上的關(guān)系。 結(jié)果: 1、 Tunel法檢測腦出血后A組凋亡細胞B組C組;統(tǒng)計學(xué)分析: A36.30%±2.83%,B22.55%±3.49%,C17.70%±2.62%,,p0.01。 2、在早期人腦出血中越靠近血腫表面p75NTR表達增加,A組p75NTR陽性細胞率B組C組(х2=15.501v=2p0.01);各組p75NTR表達與凋亡細胞率比較變化程度一致,呈正比(A組:r=0.6396,p=0.0024;B組:r=0.7889,p0.0001;C組:r=0.8772,p0.0001);TrkA在各組中有表達,但三組標(biāo)本中TrkA無差異(х2=0.877v=2p0.05),與細胞凋亡無明顯聯(lián)系;p75NTR/TrkA比率在A組中高于其余二組(F=4.851,p=0.011)。 3、p75NTR在7h組中表達明顯強于≤7h組(Z=-2.566p=0.01),在60ml組中表達亦強于60ml組(Z=-2.692p=0.007)。而TrkA在上述組中無表達差異(p0.05)。 結(jié)論: 1、HICH后腦組織中凋亡細胞由遠及近增多; 2、腦出血后p75NTR在TrkA低表達或者無活性的環(huán)境中引發(fā)細胞凋亡;越靠近血腫表面,其促凋亡作用越強; 3、TrkA在早期HICH腦組織中無表達差異,與細胞凋亡無相關(guān); 4、p75NTR/TrkA比率的變化影響出血后細胞凋亡。
[Abstract]:Aim: to investigate the expression of p75 NTRG TrkA after early hypertensive intracerebral hemorrhage, and to elucidate the relationship between p75 NTR TrkA and apoptosis.
