三氧化二砷對神經母細胞瘤細胞Trk基因家族表達的影響

【摘要】:目的神經母細胞瘤(neuroblastoma,NB)是兒童常見的實體瘤,三氧化二砷(Arsenic troxide,As_2O_3)現(xiàn)已廣泛應用于治療實體腫瘤,為了探討As_2O_3治療NB的可行性,本研究檢測As_2O_3對人SK-N-SH細胞周期及Trk基因家族表達的影響。方法通過流式細胞術檢測4μmol/L濃度的As_2O_3作用48h后SK-N-SH細胞細胞周期的變化;qRT-PCR法檢測As_2O_3在1、2和4μmol/L濃度下對SK-N-SH細胞分別作用24和48h后,對SK-N-SH細胞內TrkA、TrkB和TrkC基因表達的影響。結果用4μmol/L As_2O_3作用48h后,SK-N-SH細胞的G2/M期細胞比例為(30.93±0.34)%,而對照組G_2/M期細胞比例為(5.29±0.23)%,兩組間差異有統(tǒng)計學意義,P0.01。1μmol/L As_2O_3作用SK-N-SH細胞24h的TrkA比值為1.43±0.17,48h為3.88±0.26,兩者比較差異有明顯統(tǒng)計學意義,P0.01,但隨著As_2O_3濃度繼續(xù)增大至4μmol/L TrkA反而下調。SK-N-SH細胞的TrkB及TrkC基因表達均隨著As_2O_3濃度增加而上調,4μmol/L濃度下與對照組比較差異有統(tǒng)計學意義(P0.05),48h比24h上調更明顯。結論初步研究結果表明,As_2O_3不僅對NB的SK-N-SH細胞有明顯殺傷作用,使其阻滯在G_2/M期,還能使與NB預后密切相關的Trk基因的表達發(fā)生變化,As_2O_3有用于NB治療的潛在臨床價值。
[Abstract]:The purpose of neuroblastoma (neuroblastoma, NB) is a common solid tumor, arsenic trioxide (Arsenic troxide As_2O_3) has been widely used in the treatment of solid tumors, in order to evaluate the feasibility of As_2O_3 in the treatment of NB, this study detected the expression of As_2O_3 on SK-N-SH cell cycle and Trk gene family. The method was examined by flow cytometry. As_2O_3 48h 4 mol/L concentration after SK-N-SH cell cycle changes; detection of As_2O_3 qRT-PCR in 1, 2 and 4 mol/L concentration of SK-N-SH cells respectively 24 and 48h, on the expression of TrkA, TrkB and TrkC genes in SK-N-SH cells influence. Results after 4 48h mol/L treatment, the proportion of G2/M phase cells in SK-N-SH cells was (30.93 + 0.34)%, while in the control group, the proportion of G_2/M phase cells was (5.29 + 0.23)%. The difference between the two groups was statistically significant (P0.01). The proportion of G_2/M cells in the control group was (5.29 + 0.23)%. The TrkA ratio of 24h cells treated with 1 mu mol/L As_2O_3 was 1.43 + 0.17,48h, which was 3.88 + 0.26. The difference between them was statistically significant, P0.01, but decreased with increasing As_2O_3 concentration to 4 SK-N-SH mol/L TrkA. The expression of TrkB and TrkC genes in SK-N-SH cells increased with the increase of As_2O_3 concentration. There was a significant difference between the 4 mol/L concentration and the control group (P0.05), 48h increased more significantly than 24h. Conclusion the preliminary results indicate that As_2O_3 not only has obvious killing effect on NB SK-N-SH cells, but also blocks G_2/M phase, and also changes the expression of Trk gene, which is closely related to NB prognosis. As_2O_3 has potential clinical value for NB therapy.
