植物滅螺劑有效成分的定量及其作用機(jī)理研究

【摘要】: 血吸蟲(chóng)病是世界上僅次于瘧疾的第二大寄生蟲(chóng)病。釘螺是血吸蟲(chóng)的唯一中間宿主,消滅釘螺能有效控制血吸蟲(chóng)病的傳播。Nic是常用的化學(xué)滅螺藥物,但顯效慢、價(jià)格高、對(duì)魚(yú)毒性大。植物滅螺劑因有低毒、易生物降解、價(jià)廉等技術(shù)經(jīng)濟(jì)優(yōu)勢(shì)而受到關(guān)注。本文研究了新型植物滅螺劑(產(chǎn)品)活性成分的定量及作用機(jī)理,主要結(jié)果如下: 1.建立了HPLC法定量分析產(chǎn)品有效成分SX-Ⅰ的方法。方法:色譜柱,C185μm 4.6×150 mm;流動(dòng)相,MeOH- ACN-NH_4AC(V/V/V,30:60:10);流速,1.0mL/min;檢測(cè),UV210 nm。結(jié)果:SX-Ⅰ線性范圍為100~500μg/mL,平均回收率108.8%,變異系數(shù)(RSD)0.14%。結(jié)論:該方法簡(jiǎn)捷、準(zhǔn)確,可用于該產(chǎn)品有效成分SX-Ⅰ的質(zhì)量控制。 2.研究了NM和SX-Ⅰ滅螺藥物對(duì)釘螺酶活性及蛋白的誘導(dǎo)作用。方法:HPLC檢測(cè)及PAGE和SDS-PAGE電泳分析。結(jié)果:釘螺AchE、GOT的活性隨浸泡時(shí)間延長(zhǎng),先增強(qiáng)后減弱;而釘螺的總的酶帶數(shù)也出現(xiàn)先增加后減少的現(xiàn)象。結(jié)論:NM和SX-Ⅰ溶液滅殺釘螺,使其正常的物質(zhì)代謝平衡被破壞,蛋白合成受阻,導(dǎo)致釘螺中毒死亡。 3.測(cè)定了不同濃度SX果實(shí)粉、AC種粉及CHSO_4復(fù)配對(duì)釘螺AchE活性的影響。方法:改進(jìn)的Ellman法測(cè)定AchE對(duì)藥物的靈敏度,采用L_(25)(5~3)正交試驗(yàn)和極差分析等方法,試驗(yàn)確定3因素及其5水平,測(cè)定酶活性抑制率最高的條件組合。結(jié)果:滅螺藥物復(fù)配對(duì)釘螺AchE活性抑制率的最佳組合為:SX果實(shí)粉25mg/L、AC種粉16 mg/L、CUSO_42 mg/L。結(jié)論:該方法簡(jiǎn)單、有效,可用于研發(fā)對(duì)釘螺高毒、魚(yú)低毒的新型滅螺劑提供參考。
