【摘要】：目的:研究生長激素對正畸牙齒移動過程中牙根周圍組織中骨橋蛋白(osteopontin,OPN)表達(dá)的影響。方法:將40只2月齡體重約200 g雄性Wistar大鼠隨機(jī)分成對照組和實驗組,每組各20只。建立大鼠上頜第一磨牙正畸牙齒移動模型。實驗組大鼠予0.15 U/(kg.d)的生長激素腹部皮下注射;對照組大鼠每天注射等量的生理鹽水。于大鼠上頜中切牙和左側(cè)上頜第一磨牙之間放置鎳鈦拉簧,拉力為0.49 N,使磨牙向中切牙方向移動。加力后的第3,7,10,14天,用心臟灌注法處死所有大鼠。取下左側(cè)上頜骨,制備第一磨牙近遠(yuǎn)中向的縱向切片,免疫組織化學(xué)染色觀察近頰根與遠(yuǎn)頰根之間牙槽骨的壓力區(qū)中牙周膜OPN的表達(dá),并進(jìn)行半定量分析。結(jié)果:實驗組和對照組的成骨細(xì)胞、破骨細(xì)胞和骨細(xì)胞的胞質(zhì)中都有OPN的陽性表達(dá),并且有相同的變化趨勢,OPN陽性表達(dá)早期隨破骨細(xì)胞的增多而增加,后期隨破骨細(xì)胞的減少而降低。正畸加力后第7天,與對照組相比,實驗組OPN陽性表達(dá)更強(P0.05);第10天,與對照組相比,實驗組OPN陽性表達(dá)下降更快,實驗組明顯低于對照組(P0.05)。結(jié)論:全身應(yīng)用生長激素能影響大鼠移動牙齒時牙周組織中OPN的表達(dá),可能對正畸牙齒移動過程中壓力側(cè)骨的吸收發(fā)揮重要的調(diào)節(jié)作用。
[Abstract]:Aim: to investigate the effect of long hormone on the expression of osteopontin (osteopontin,OPN) in periroot tissue during orthodontic tooth movement. Methods: forty 2-month-old male Wistar rats were randomly divided into two groups: control group (n = 20) and experimental group (n = 20). The orthodontic tooth movement model of maxillary first molar was established in rats. The rats in the experimental group were subcutaneously injected with 0.15U / (kg.d) of growth hormone, and the rats in the control group were injected with the same amount of saline every day. A Ni-Ti pull spring was placed between the maxillary middle incisor and the left maxillary first molar. The tensile force was 0.49 N, which made the molar move toward the middle incisor. At days 3, 7, 10, 14, all rats were killed by cardiac perfusion. The left maxillary bone was removed and the proximal and distal longitudinal sections of the first molar were prepared. Immunohistochemical staining was used to observe the expression of periodontal ligament OPN in the pressure region of the alveolar bone between the proximal and distal buccal roots, and semi-quantitative analysis was carried out. Results: there was positive expression of OPN in osteoblasts, osteoclasts and osteoblasts in both experimental and control groups. The expression of OPN in osteoblasts, osteoclasts and osteoblasts in the early stage increased with the increase of osteoclasts. At the later stage, the number of osteoclasts decreased with the decrease of osteoclasts. On the 7th day after orthodontic treatment, the positive expression of OPN in the experimental group was stronger than that in the control group (P0.05); on the 10th day, the positive expression of OPN in the experimental group decreased faster than that in the control group, and was significantly lower in the experimental group than that in the control group (P0.05). Conclusion: growth hormone can affect the expression of OPN in periodontal tissue during tooth movement in rats, and may play an important role in regulating the bone resorption of pressure side during orthodontic tooth movement.
