【摘要】：目的： 探討乳桿菌A-2代謝產(chǎn)物對(duì)人舌鱗癌CAL-27細(xì)胞的增殖抑制與誘導(dǎo)凋亡的作用。 方法： 乳桿菌A-2分別經(jīng)復(fù)原乳清培養(yǎng)基和復(fù)原乳清菊糖培養(yǎng)基培養(yǎng)，制備相應(yīng)的代謝產(chǎn)物1（LSPM1）和代謝產(chǎn)物2(LSPM2)；采用MTT法檢測(cè)對(duì)CAL-27細(xì)胞的體外增殖抑制作用；通過(guò)吖啶橙染色，熒光顯微鏡下觀察細(xì)胞形態(tài)學(xué)改變；采用流式細(xì)胞儀和DNA瓊脂凝膠電泳法檢測(cè)細(xì)胞凋亡情況。 結(jié)果： 在LSPM1和LSPM2在3.0mg/mL~48mg/mL的區(qū)間內(nèi)，隨著代謝產(chǎn)物濃度的增加，對(duì)CAL-27細(xì)胞的抑制作用逐漸增強(qiáng)。LSPM1和LSPM2的半抑制濃度(IC50)分別為15.1mg/mL和10.27mg/mL；熒光顯微鏡下觀察，凋亡細(xì)胞的細(xì)胞核不規(guī)則，細(xì)胞皺縮，，凝聚呈新月形或馬蹄形，貼壁不良；流式細(xì)胞儀結(jié)果顯示：經(jīng)LSPM1和LSPM2處理的CAL-27細(xì)胞凋亡率均高于空白對(duì)照組；瓊脂糖凝膠電泳顯示出典型的DNA梯形。 結(jié)論： 乳桿菌A-2代謝產(chǎn)物可以在體外抑制人舌鱗癌CAL-27細(xì)胞生長(zhǎng)，誘導(dǎo)CAL-27細(xì)胞凋亡。
[Abstract]:Aim: to investigate the inhibitory effect of Lactobacillus A-2 metabolite on proliferation and apoptosis of human tongue squamous cell carcinoma (CAL-27) cells. Methods: Lactobacillus A-2 was cultured on restoration whey medium and restored whey inulin medium respectively to prepare metabolite 1 (LSPM1) and metabolite 2 (LSPM2). Cell morphology was observed by acridine orange staining, and apoptosis was detected by flow cytometry and DNA agarose gel electrophoresis. Results: in the 3.0mg/mL~48mg/mL region of LSPM1 and LSPM2, with the increase of the concentration of metabolites, the inhibitory effect on CAL-27 cells was gradually enhanced. The half inhibitory concentrations (IC50) of LSPM1 and LSPM2 were 15.1mg/mL and 10.27 mg / mL, respectively. The apoptotic cells showed irregular nuclei, crumpled cells, crescent or horseshoe shape, and poor adhesion. The results of flow cytometry showed that the apoptotic rate of CAL-27 cells treated with LSPM1 and LSPM2 was higher than that of the control group. Agarose gel electrophoresis showed a typical DNA trapezoid. Conclusion: Lactobacillus A-2 metabolites can inhibit the growth of human tongue squamous cell carcinoma (CAL-27) cells and induce apoptosis of CAL-27 cells in vitro.
