人臍帶Wharton’s Jelly來源間充質(zhì)干細(xì)胞與人牙周膜干細(xì)胞成骨分化能力的對(duì)比研究

【摘要】:目的:比較人臍帶Wharton’s Jelly來源間充質(zhì)干細(xì)胞(human umbilical cord Wharton’s Jelly-derived mesechymal stem cells,h UCWJMSCs)與人牙周膜干細(xì)胞(periodontal mesenchymal stem cells,h PDLSCs)成骨分化能力。方法:體外培養(yǎng)h UCWJMSCs和h PDLSCs。MTT法檢測(cè)細(xì)胞增殖情況;成骨誘導(dǎo)后測(cè)定細(xì)胞的ALP活性,茜素紅染色檢測(cè)細(xì)胞礦化能力,Real-time PCR分析OPN和Runx2基因的表達(dá)。結(jié)果:hUCWJMSCs增殖能力高于hPDLSCs;經(jīng)礦化誘導(dǎo)后hPDLSCsALP表達(dá)、礦化結(jié)節(jié)形成高于hUCWJMSCs(P0.05);Runx2在hPDLSCs中表達(dá)高于hUCWJMSCs(P0.05);而hUCWJMSCs中OPN表達(dá)高于hPDLSCs(P0.05)。結(jié)論:h UCWJMSCs、hPDLSCs均具有成骨分化能力,hPDLSCs成骨分化能力較強(qiáng)。
[Abstract]:Aim: to compare the osteogenic differentiation between human umbilical cord Wharton's Jelly derived mesenchymal stem cells (human umbilical cord Wharton's Jelly-derived mesechymal stem cells,h UCWJMSCs) and human periodontal ligament stem cells (periodontal mesenchymal stem cells,h PDLSCs). Methods: the cell proliferation was detected by h UCWJMSCs and h PDLSCs.MTT in vitro, the activity of ALP was measured after osteogenesis induction, and the mineralization ability of cells was detected by alizarin red staining. The expression of OPN and Runx2 genes was detected by Real-time PCR. Results the proliferative ability of hPDLSCs; was higher than that of hPDLSCsALP induced by hPDLSCs;, and the expression of hUCWJMSCs (P0.05) Runx2 in hPDLSCs was higher than that of hUCWJMSCs (P0.05), while the expression of OPN in hUCWJMSCs was higher than that in hPDLSCs (P0.05). Conclusion all UCWJMSCs,hPDLSCs have the ability of osteogenic differentiation and the osteogenic differentiation ability of hPDLSCs is stronger than that of hPDLSCs.
