【摘要】：目的前列腺素E2(prostaglandin E2,PGE2)可以通過促進(jìn)癌細(xì)胞的增殖、抑制癌細(xì)胞的凋亡、促進(jìn)癌細(xì)胞的轉(zhuǎn)移等方式促進(jìn)口腔鱗癌的發(fā)生發(fā)展,但關(guān)于其具體作用機(jī)制罕見報(bào)道。因此,本研究旨在檢測(cè)口腔鱗狀細(xì)胞癌組織和相應(yīng)癌旁組織中花生四烯酸(arachidonic acid,AA)及前列腺素E2受體3亞型(E-prostanoid 3,EP3)的表達(dá)。方法選取南方醫(yī)科大學(xué)南方醫(yī)院2013-05-01-2015-05-01行手術(shù)治療的口腔鱗癌患者40例,將口腔鱗狀細(xì)胞癌組織作為觀察組(40例),相應(yīng)癌旁組織作為對(duì)照組(40例)。ELISA方法檢測(cè)AA的表達(dá),Real-time PCR方法檢測(cè)EP3受體mRNA的表達(dá),免疫組織化學(xué)染色方法檢測(cè)EP3受體蛋白的表達(dá)。結(jié)果與癌旁組織相比,AA在口腔鱗狀細(xì)胞癌組織中的表達(dá)明顯升高。40例口腔鱗癌組織中AA的平均質(zhì)量分?jǐn)?shù)為(477.4±39.84)pg/g,40例癌旁組織中AA的平均質(zhì)量分?jǐn)?shù)為(38.4±4.92)pg/g,差異有統(tǒng)計(jì)學(xué)意義,t=3.146,P=0.001。與癌旁組織相比,鱗癌組織中EP3受體mRNA的表達(dá)明顯降低。40例鱗癌組織EP3受體mRNA的相對(duì)平均表達(dá)量為0.43±0.15,差異有統(tǒng)計(jì)學(xué)意義,t=2.365,P=0.011。與癌旁組織相比,鱗癌組織中EP3受體蛋白的表達(dá)明顯降低,20例癌旁組織呈陽性表達(dá),而18例鱗癌組織呈弱陽性表達(dá),差異有統(tǒng)計(jì)學(xué)意義,T=-114,P=0.016。結(jié)論口腔鱗狀細(xì)胞癌組織中,AA表達(dá)量的顯著上調(diào),EP3受體表達(dá)量的顯著下調(diào),可能在口腔鱗狀細(xì)胞癌的發(fā)生發(fā)展過程中發(fā)揮了重要作用。
[Abstract]:Objective prostaglandin E 2 (prostaglandin E 2) can promote the development of oral squamous cell carcinoma by promoting the proliferation of cancer cells, inhibiting the apoptosis of cancer cells and promoting the metastasis of cancer cells. Therefore, the purpose of this study was to detect the expression of arachidonic acid (arachidonic acid,AA) and prostaglandin E 2 receptor 3 subtype (E-prostanoid 3P 3) in oral squamous cell carcinoma and adjacent tissues. Methods Forty patients with oral squamous cell carcinoma (OSCC) who underwent surgical treatment from 2013-05-01-2015-05-01 in Southern Hospital of Southern Medical University were selected. Oral squamous cell carcinoma (OSCC) tissues were used as observation group (n = 40) and adjacent tissues as control group (n = 40). Elisa was used to detect the expression of AA. Real-time PCR was used to detect the expression of EP3 receptor mRNA, and immunohistochemical staining was used to detect the expression of EP3 receptor protein. Results the expression of AA in oral squamous cell carcinoma tissues was significantly higher than that in adjacent tissues. The mean mass fraction of AA in 40 cases of oral squamous cell carcinoma was (477.4 鹵39.84) pg/g,40. The mean mass fraction of AA in adjacent tissues was (38.4 鹵4.92) pg/g,. The expression of EP3 receptor mRNA in squamous cell carcinoma was significantly lower than that in adjacent tissues. The relative average expression of EP3 receptor mRNA in 40 cases of squamous cell carcinoma was 0.43 鹵0.15, and the difference was statistically significant. Compared with adjacent tissues, the expression of EP3 receptor protein in squamous cell carcinoma decreased significantly in 20 cases of squamous cell carcinoma, while the expression of EP3 receptor protein in 18 cases of squamous cell carcinoma was weakly positive. The difference was statistically significant (P 0.016). Conclusion the upregulation of AA expression in oral squamous cell carcinoma may play an important role in the development of oral squamous cell carcinoma.
