【摘要】：目的:觀察抗炎中藥混合制劑Zyflamend(ZF)對(duì)DMBA誘發(fā)的金黃地鼠口腔癌的抑制作用。方法:116只金黃地鼠,10只設(shè)為陰性對(duì)照,其余用0.5%的DMBA石蠟油涂于地鼠左側(cè)頰囊,3次/周,共涂6周。從第7周開(kāi)始,將動(dòng)物分為3組。給低濃度組(n=33)地鼠左側(cè)頰囊局部涂抹1∶1石蠟油稀釋的ZF,高濃度組(n=33)涂ZF原液,3次/周,連續(xù)18周,陽(yáng)性對(duì)照組(n=40)不給藥。第24周末處死動(dòng)物,處死前2 h腹腔注射50 mg/kg的5-溴脫氧尿嘧啶(BrdU)。取左側(cè)頰囊進(jìn)行組織病理學(xué)觀察和BrdU免疫組化染色。結(jié)果:未防治組低、高濃度ZF組,肉眼觀察腫瘤發(fā)生率分別為86.7%、53.6%(P0.01)和50.0%(P0.01);組織病理學(xué)觀察鱗狀細(xì)胞癌的發(fā)病率分別為86.7%、78.6%(P0.05)和64.3%(P0.05)。ZF高濃度組的病變中組織,BrdU的增殖指數(shù)顯著降低(P0.05)。結(jié)論:Zyflamend能夠降低DMBA誘導(dǎo)金黃地鼠口腔癌的發(fā)生率,其機(jī)制可能與抑制細(xì)胞增殖有關(guān)。
[Abstract]:Aim: to observe the inhibitory effect of Zyflamendment ZF on oral cancer induced by DMBA in hamsters. Methods 10 golden hamsters were used as negative control, and the rest were coated with 0.5% DMBA paraffin oil on the left cheek pouch of hamster three times a week for 6 weeks. From week 7, animals were divided into 3 groups. The left cheek pouch of the hamster was locally smeared with 1:1 paraffin oil diluted ZF, and the high concentration group was treated with ZF solution 3 times a week for 18 weeks. Animals were killed at the end of the 24th week, and 50 mg/kg 5-bromodeoxyuridine (BrdU) was injected intraperitoneally 2 hours before death. The left cheek pouch was taken for histopathological observation and BrdU immunohistochemical staining. Results: the incidences of tumor were 86.7% (P 0.01) and 50.01% (P 0.01), respectively, and the incidences of histopathological observation were 86.7% (P 0.05) and 78.6% (P 0.05), respectively, and the proliferative index of BrdU was significantly decreased in the group of 64.3%(P0.05).ZF with high concentration of ZF (86.7%) and the group of 64.3%(P0.05).ZF with high concentration (P 0.01), and the incidence of histopathological observation of squamous cell carcinoma was 86.7% (P 0.05) and the proliferative index of BrdU was significantly lower than that of 64.3%(P0.05).ZF group (P 0.05). ConclusionZyflamend can reduce the incidence of oral cancer induced by DMBA in golden hamsters, and its mechanism may be related to the inhibition of cell proliferation.
