3種擬癡呆動物模型在Morris水迷宮行為學(xué)測試中學(xué)習(xí)記憶行為的差異

【摘要】:目的 Morris水迷宮實驗是一種評價空間學(xué)習(xí)記憶能力的經(jīng)典方法,是評價癡呆動物模型復(fù)制結(jié)果的客觀指標(biāo)。近年來癡呆動物模型已成為研究老年性癡呆的重要手段。文中評價3種常見擬癡呆動物模型在Morris水迷宮行為學(xué)測試中學(xué)習(xí)記憶行為的差異。方法分別使用SD大鼠、昆明種小鼠以及快速老化小鼠制備Aβ腦室注射致癡呆模型、東莨菪堿腹腔注射致記憶障礙模型、快速老化模型。采用Morris水迷宮行為學(xué)測試方法,將這3種模型通過與其相應(yīng)的對照組比較,考察其行為學(xué)差異。結(jié)果東莨菪堿模型與快速老化模型逃避潛伏期時間短于相應(yīng)對照組,其中以東莨菪堿模型差異尤為顯著[(35.47±3.78)s vs(50.61±3.94)s,P0.01];快速老化組模型與對照組差異有統(tǒng)計學(xué)意義[(17.39±4.71)s vs(33.79±7.81)s,P0.05];Aβ致癡呆模型與其對照組比較,模型定位航行實驗成績差異無統(tǒng)計學(xué)意義(P0.05)。結(jié)論東莨菪堿腹腔注射致記憶障礙模型較其他2種常用模型更為理想。Morris水迷宮行為學(xué)測試方法是一種評價擬癡呆動物的理想方法。
[Abstract]:Objective Morris water maze test is a classical method to evaluate spatial learning and memory ability, and it is an objective index to evaluate the replicating results of dementia animal models. In recent years, animal models of dementia have become an important means to study Alzheimer's disease. The differences of learning and memory behaviors of three common dementia animal models in Morris water maze behavior test were evaluated in this paper. Methods SD rats, Kunming mice and rapid aging mice were used to establish dementia model induced by A 尾 intracerebroventricular injection, amnesia induced by scopolamine intraperitoneal injection, and rapid aging model. The behavioral differences of the three models were compared with those of the control group by Morris water maze test method. Results the escape latency of scopolamine model and rapid aging model was shorter than that of control group. There were significant differences in scopolamine model (35.47 鹵3.78s vs(50.61 鹵3.94 vs(50.61 P0.01) and rapid aging group (17.39 鹵4.71s vs(33.79 鹵7.81 vs(33.79 鹵7.81g / kg) compared with control group (P 0.05). Conclusion the amnesia model induced by scopolamine intraperitoneal injection is more ideal than the other two models. Morris water maze is an ideal method to evaluate dementia animals.
