【摘要】：目的:探討鹽酸米諾環(huán)素、梔子苷單獨(dú)及聯(lián)合應(yīng)用對(duì)NO誘導(dǎo)PC12細(xì)胞凋亡的影響及其可能機(jī)制。方法:PC12細(xì)胞分為對(duì)照組、鹽酸米諾環(huán)素組、梔子苷組和鹽酸米諾環(huán)素+梔子苷組(聯(lián)合應(yīng)用組)。4組均用終濃度為0.5 mmol/L的NO供體SIN-1誘導(dǎo)細(xì)胞凋亡,SIN-1干預(yù)前12 h鹽酸米諾環(huán)素組加入終濃度為0.1 mmol/L的鹽酸米諾環(huán)素,梔子苷組加入終濃度為0.25 mmol/L的梔子苷,聯(lián)合應(yīng)用組加入同上濃度的鹽酸米諾環(huán)素和梔子苷,對(duì)照組不給藥。CCK-8法測(cè)定細(xì)胞存活率,Hoechest 33342熒光染色觀察細(xì)胞凋亡情況,流式細(xì)胞儀測(cè)定細(xì)胞凋亡率,Western blot測(cè)定凋亡相關(guān)蛋白Caspase-3和Bcl-2的表達(dá)。結(jié)果:與對(duì)照組相比,單用鹽酸米諾環(huán)素或梔子苷均能提高NO誘導(dǎo)的PC12細(xì)胞存活率,降低細(xì)胞凋亡率,增加Bcl-2蛋白表達(dá),減少Caspase-3蛋白表達(dá)(P均0.05);二者聯(lián)合應(yīng)用對(duì)上述指標(biāo)的影響具有協(xié)同效應(yīng)(P0.05)。結(jié)論:鹽酸米諾環(huán)素和梔子苷均能抑制PC12細(xì)胞凋亡,且二者聯(lián)合應(yīng)用作用優(yōu)于單獨(dú)應(yīng)用,其機(jī)制可能與抑制Caspase-3依賴(lài)性凋亡信號(hào)通路有關(guān)。
[Abstract]:Aim: to investigate the effect of minocycline hydrochloride and geniposide on apoptosis of PC12 cells induced by no and its possible mechanism. Methods Twenty PC12 cells were divided into control group and minocycline hydrochloride group. Both geniposide group and minocycline geniposide hydrochloride group (combined with minocycline hydrochloride group) were treated with no donor SIN-1 with final concentration of 0.5 mmol/L to induce apoptosis 12 hours before intervention with minocycline hydrochloride and minocycline hydrochloride with final concentration of 0.1 mmol/L. The geniposide group was added with 33342 mmol/L geniposide, the same concentration of minocycline hydrochloride and geniposide was added in the combination group, and the cell apoptosis was observed by Hoechest 33342 fluorescence staining without CCK-8 in the control group. Apoptosis rate was determined by flow cytometry and the expression of apoptosis-related protein Caspase-3 and Bcl-2 was detected by Western blot. Results: compared with the control group, minocycline hydrochloride or geniposide alone could increase the survival rate of PC12 cells induced by no, decrease the rate of apoptosis and increase the expression of Bcl-2 protein. The combined use of both of them had a synergistic effect on the expression of Caspase-3 protein (P 0.05). Conclusion: minocycline hydrochloride and geniposide can inhibit the apoptosis of PC12 cells, and their combined effect is better than that of single application. The mechanism may be related to the inhibition of Caspase-3 dependent apoptosis signaling pathway.
