精神分裂癥circRNA的差異表達(dá)及其與癥狀的關(guān)系

【摘要】:目的探討精神分裂癥circRNA表達(dá)水平及其與患者陰性、陽(yáng)性癥狀的關(guān)系。方法分別選取5例精神分裂癥患者和正常人外周血進(jìn)行基因芯片篩查,確定9種存在差異表達(dá)的circRNA,并在102例精神分裂癥患者和103例正常人中進(jìn)行PCR驗(yàn)證,進(jìn)一步行精神分裂癥陽(yáng)性和陰性癥狀量表評(píng)估。結(jié)果精神分裂癥患者外周血circRNA_102101、circRNA_102315、circRNA_104597、circRNA_101835、circRNA_101836的表達(dá)水平明顯下調(diào)(P0.05或P0.01)。circRNA_103102、circRNA_103704的表達(dá)水平明顯上調(diào)(P0.01)。circRNA_102101、circRNA_103102的ΔCt值與陽(yáng)性癥狀呈明顯正相關(guān)(P0.01或P0.05)。circRNA_103704與陽(yáng)性癥狀、一般精神病理呈明顯正相關(guān)(P0.01或P0.05)。思維障礙與circRNA_102101、circRNA_103102、circRNA_102315、circRNA_103704、circRNA_102802的ΔCt值呈明顯正相關(guān)(P0.01或P0.05)。激活性與circRNA_102101、circRNA_103102、circRNA_104597、circRNA_103704、circRNA_102802的ΔCt值呈明顯正相關(guān)(P0.01或P0.05)。偏執(zhí)與circRNA_102101、circRNA_103102、ci rcRNA_103704、ci rcRNA_102802的ΔCt值呈明顯正相關(guān)(P0.01或P0.05)。攻擊性與ci rcRNA_102101、circRNA_103102、circRNA_103704、circRNA_102802的ΔCt值呈明顯正相關(guān)(P0.01或P0.05)。circRNA_103704進(jìn)入陽(yáng)性癥狀的回歸方程(P0.01);circRNA_103704、circRNA_102315進(jìn)入一般病理表現(xiàn)的回歸方程(P0.01或P0.05)。結(jié)論circRNA_103704、circRNA_103102與精神分裂癥陰性、陽(yáng)性癥狀有明顯關(guān)聯(lián),可能是精神分裂癥病理過程主導(dǎo)的調(diào)控因子。
[Abstract]:Objective to investigate the expression of circRNA in schizophrenia and its relationship with negative and positive symptoms. Methods five patients with schizophrenia and 10 healthy controls were selected for gene microarray screening, and 9 differentially expressed circRNAs were identified. PCR was performed in 102 schizophrenic patients and 103 normal controls. The scale of positive and negative symptoms of schizophrenia was further evaluated. Results the expression level of circRNA102101 circRNA1022315 circRNA104597 circRNA101836 in peripheral blood of schizophrenic patients was significantly down-regulated, P0.01n.circRNA103102circRNA103704 or P0.01circRNA102circRNA103704 was significantly up-regulated. The 螖 Ct value of 102101circRNA103102 was positively correlated with the positive symptoms (P0.01 or P0.05).circRNA_103704), and the general psychopathology was positively correlated with the symptoms (P0.01 or P0.05). There was a significant positive correlation between thinking disorder and circRNA102101 circRNAs, 103102 circRNAs, 102315 circRNAs, 103704 circRNA102802, P0.01 or P0.05. Activation was positively correlated with the 螖 Ct value of circRNAs 1031021 and circRNAs 103102 / circRNAs in 103704 circRNAs, and the 螖 Ct values of circRNA102802 were positively correlated with the number of circRNAs. The 螖 Ct value of 103102ci rcRNA103704 rcRNA_102802 was positively correlated with the 螖 Ct value of 102101 circRNAs (P0.01 or P0.05). There was a significant positive correlation between the 螖 Ct values of the tip103704 circRNA102802 and the ci rcRNA102101 circRNAs. There was a significant positive correlation between the 螖 Ct values of P0.01 or P0.05).circRNA_103704 entering the positive symptoms regression equation. P0.01 or P0.05. the number of circRNAs 103704 circRNA102315 entered the regression equation P0.01 or P0.05 for the general pathological manifestations. Conclusion\\\
